Ascocotyle longa is parasitic trematode with wide distribution throughout America, Europe, Africa, and Middle East. Despite the fact that this fish-borne pathogen has been considered an agent of human heterophyiasis in Brazil, the molecules involved in the host-parasite interaction remain unknown. The present study reports the proteome profile of A. longa metacercariae collected from the fish Mugil liza from Brazil. This infective stage for humans, mammals and birds was analyzed using nLC-MS/MS approach. We identified a large repertoire of proteins, which are mainly involved in energy metabolism and cell structure. Peptidases and immunogenic proteins were also identified, which might play roles in host-parasite interface. Our data provided unprecedented insights into the biology of A. longa and represent a first step to understand the natural host-parasite interaction. Moreover, as the first proteome characterized in this trematode, it will provide an important resource for future studies.

In recent years, the consumption of fish products has surged in European countries, being an essential part of a healthy diet. Despite representing a small part of EU production, freshwater fisheries hold considerable significance for lake-dwelling populations and tourists seeking traditional dishes. This increased fish consumption has brought to light potential health risks associated with fish-borne zoonotic helminths (FBZHs), now acknowledged as global food-borne parasites. Fish-borne zoonotic helminths belong to various taxonomic groups, including nematodes (Anisakidae), trematodes (Opisthorchiidae and Heterophyidae), and cestodes (Diphyllobothriidae). More than 50 species of FBZH are known to cause human infections, derived from eating raw or undercooked aquatic foods containing viable parasites. Despite increased attention, FBZHs remain relatively neglected compared to other food-borne pathogens due to factors like chronic disease progression and under-diagnosis. This systematic review concentrates on the prevalence of six freshwater FBZHs (, , , , , and ) in Italy and neighbouring countries. The study explores the expansion of these parasites, analysing their biological and epidemiological aspects, and the factors that influence their proliferation, such as the increased cormorant population and the lake eutrophication phenomena. In summary, this research highlights the necessity for further research, the development of spatial databases, and the establishment of a unified European policy to effectively manage these multifaceted health concerns. It strongly advocates adopting a One-Health approach to address the growing incidence of parasitic zoonoses within the context of food safety in EU countries.

Wild canides have a high epizootiological - epidemiological significance, considering that they are hosts for some parasites which spread vector born diseases. Increased frequency of certain interactions between domestic and wild canides increases the risk of occurrence, spreading and maintaining the infection of parasitic etiology in domestic canides. The research was conducted in 232 wild canides (172 red foxes and 60 golden jackals). The examined material was sampled from foxes and jackals, which were hunted down between 2010 and 2014, from 8 epizootiological areas of Serbia (North-Bačka, West-Bačka, Southern-Banat, Moravički, Zlatiborski, Raški, Rasinski and Zaječarski district). On completing the parasitological dissection and the coprological diagnostics, in wild canides protozoa from the genus Isospora were identified, 3 species of trematoda (Alaria alata, Pseudamphistomum truncatum and Metagonimus yokogawai), cestods from the genus Taenia and 5 species of nematodes (Toxocara canis, Ancylostomatidae, Trichuris vulpis and Capillaria aerophila). The finding of M. yokogawai in golden jackals were, to the best of our knowledge, one of the first diagnosed cases of metagonimosis in golden jackals in Serbia. The continued monitoring of the parasitic fauna of wild canides is needed to establish the widespread of the zoonoses in different regions of Serbia, because they present the reservoirs and/or sources of these infections.

Histopathological diagnosis was used to understand the pathological events associated with Heterophyes heterophyes (H. heterophyes) infection. CD3 and CD79α antibodies had been used as markers for both T and. B lymphocytes respectively. Immunohistochemical techniques had several advantages as remarkable sensitivity and specificity. This study aims to evaluate the roles-of praziquantel (PZQ) and aminoguanidine (AG) treatment in H heterophyes infected dogs pathologically and immunohisto-chemically. Study design included experimental infection of dogs with encysted metacercariae of H heterophyes followed by treatment with PZQ and AG. Tissue samples were taken from small intestinal, liver, heart and lung of all groups for histopathological and immunohistochemical studies. Pathological changes were detected in infected tissues by histopathological examination. There was different degree of CD79α+B lymphocytic & CD3+T lymphocytic infiltration detected in immuno-histochemical stained tissues. PZQ caused improvement of pathological changes in the small intestine. However the cellular inflammatory infiltration increased. There was reduction in inflammatory infiltration after intake of AG. Both PZQ and AG improved the pathological changes in the.liver, heart and lung, while the cellular inflammatory infiltration increased after PZQ and reduced by AG. Moreover in the lung AG improves pulmonary congestion and alveolar wall thickness.

In this study of Metagonimus suifunensis (M. suifunensis) in the Russian Southern Far East, the variability of the full-length sequences of the cytochrome b (cytb) mtDNA gene was assessed for the first time. In addition, the cox1 mtDNA gene sequences were also obtained for this species from new localities. In total, 87 and 81 sequences of the cytb and cox1 genes, respectively, were used in the current study. The cytb gene proved more promising and revealed two haplogroups that are associated with the spatial distribution of the species: geographical isolation caused the fixation of differences between northern and southern populations. In addition, the results obtained for the cytb gene opened up new perspectives in the analysis of sequences of the cox1 gene, which was not sufficiently effective as a sole marker. Based on data for both mitochondrial genes, molecular processes influencing the formation of the modern population were analysed for M. suifunensis. The new data confirmed the previously expressed opinion that this species colonized the study territory from north to south and will form the basis for determining possible ways of its further expansion, which is important for predicting the emergence of new foci of metagonimosis.

Seven laboratory mammal and bird species were orally inoculated with 200-1,000 encysted Metagonimus hakubaensis metacercariae that had been isolated from naturally infected lampreys (Lethenteron reissneri) captured in Aomori Prefecture. At 8 and 15 days post-infection, adult flukes were recovered from all of the laboratory animals tested, and therefore, hamster, rat, mouse, dog, cat, chicken and quail were considered as final hosts of M. hakubaensis. Recovery rates of the fluke were higher in dogs and hamsters than in cats, rats, mice, chickens and quails. The flukes recovered from dogs and hamsters showed increased body length and higher fecundity than those recovered from the other hosts. These results indicate that the suitability of dogs and hamsters for M. hakubaensis infection is higher than that of the other laboratory animals.

This study reports the presence of cartilaginous cysts in zebrafish gills, purchased at the end of 2012 before the application of the Italian Legislative Decree 26/2014 and used for experimental research. It highlights the importance of using specific pathogen-free organisms from reliable production establishments to avoid obtaining false results in experiments. Of 20 samples analyzed, 4 showed cysts in gills, with abnormal tissue modification and cartilaginous metaplasia induced by prolonged parasitic insult. The results obtained from the periodic health monitoring program showed the presence of metacercariae of digenean trematodes, specifically identified as .

This study was conducted to investigate the prevalence of intestinal parasites among students from Goseonggun, Gangwon-do and Hadong-gun, Gyeongsangnam-do as typical low and high endemic counties. From May to July 2017, a total of 2,033 fecal samples were collected at 27 elementary, 10 junior high, and 8 high schools from 2 counties and examined by the Kato-Katz technique for egg-positive surveys (Collection rate: 37.02% [2,033/5,492]). Of the participants examined, 13 (0.64%) were found to harbor eggs of 3 parasitic species, Trichuris trichiura, Clonorchis sinensis and Metagonimus yokogawai. Based on the regional distribution, the egg-positive rate in Goseong-gun was 0% (0/550) and that in Hadong-gun was 0.88% (13/1,483). The positive rates for C. sinensis, M. yokogawai, and T. trichiura in Hadonggun were 0.20% (3/1,483), 0.61% (9/1,483), and 0.07% (1/1,483), respectively. The present survey showed that the prevalence of parasitic infection among students is currently very low even in remote, previously endemic areas, and the present status of parasitic diseases can be summarized as some transmission of fish/food-borne trematodes. Thus, it is necessary to carefully, continuously monitor the trematode infection status, particularly of C. sinensis and M. yokogawai.

Freshwater snails play an essential role in the transmission of trematode parasitic flatworms that can infect wild and domestic animals, as well as humans. This study aimed to investigate the rate of cercarial infections in freshwater snails collected from two study areas, Inlay Lake and Yezin Dam, in Myanmar. A total of 4,740 snail samples were collected from Inlay Lake (n = 3,837) and Yezin Dam (n = 903), and infection rate by cercarial emergence was examined. Cercarial DNA samples were analysed by PCR. Based on morphological characteristics, eleven snail species and eight cercarial types were identified. Snails of Melanoides tuberculata in the family Thiaridae were found as the most abundant, followed by Indoplanorbis exustus of the family Planorbidae, in both study areas. The infection rate by cercarial emergence in snails in Inlay Lake and Yezin Dam was 5.8% (224/3,837) and 48.6% (439/903), respectively. Echinostome cercariae showed the highest infection rate in both study areas. Phylogenetic analysis of cercarial internal transcribed spacer 2 (ITS2) sequences revealed that at least seven cercaria types belonged to five digenean trematode families, two of which were zoonotic trematodes in the families of Opisthorchiidae/Heterophyidae and Schistosomatidae. Furthermore, cercarial 28S ribosomal RNA gene analysis showed that the furcocercous cercariae in Yezin Dam were identified as Schistosoma spindale, a causative agent of ruminant schistosomiasis. This is the first report on zoonotic trematode cercariae in snails in Myanmar. The findings indicate that various snail species act as intermediate host for trematode species that infect aquatic animals, mammals and humans in the country.

The assassin snail genus is widespread in Southeast Asia, and is distributed all over the world via the aquarium trade. One species of genus , , is known to act as intermediate host of parasitic trematodes. This study investigates the taxonomic diversity of larval trematodes infecting . and in Thailand. Larval trematodes were identified by combining morphological and DNA sequence data (cytochrome oxidase I and internal transcribed spacer 2). Species delimitation methods were used to explore larval trematode species boundaries. A total of 1107 specimens of sp. were collected from 25 localities in Thailand. Sixty-two specimens of . ( = 33) and . ( = 29) were infected by zoogonid cercariae, heterophyid metacercariae and echinostome metacercariae, with an overall prevalence of 5.6% (62/1107) and population-level prevalences in the range of 0.0-22.3%. DNA sequence data confirmed that the larval trematodes belong to the families Echinostomatidae, Heterophyidae and Zoogonidae. As such, this is the first report of zoogonid cercariae and heterophyid metacercariae in . , and echinostome metacercariae in . . Moreover, this study provides evidence of tentative species-level differentiation between Thai sp. and Cambodian , as well as within , and .
