Use of phorate as a broad spectrum pesticide in agricultural crops is finding disfavor due to persistence of both the principal compound as well as its toxic residues in soil. Three phorate utilizing bacterial species (Pseudomonas sp. strain Imbl 4.3, Pseudomonas sp. strain Imbl 5.1, Pseudomonas sp. strain Imbl 5.2) were isolated from field soils. Comparative phorate degradation analysis of these species in liquid cultures identified Pseudomonas sp. strain Imbl 5.1 to cause complete metabolization of phorate during seven days as compared to the other two species in 13 days. In soils amended with phorate at different levels (100, 200, 300 mg kg(-1) soil), Pseudomonas sp. strain Imbl 5.1 resulted in active metabolization of phorate by between 94.66% and 95.62% establishing the same to be a potent bacterium for significantly relieving soil from phorate residues. Metabolization of phorate to these phorate residues did not follow the first order kinetics. This study proves that Pseudomonas sp. strain Imbl 5.1 has huge potential for active bioremediation of phorate both in liquid cultures and agricultural soils.

This work performed a large scale assessment for organophosphorus pesticides (OPPs) degradation activity of 121 (.) strains. Six strains (P9, IMAU80110, IMAU40100, IMAU10585, IMAU10209, and IMAU80070) were found to possess high capacity of degrading three commonly used OPPs, namely dimethoate, phorate, and omethoate; and they were selected for more detailed characterization. Moreover, the three OPPs were mainly detected in the culture supernatants but not in the cell extracts, further confirming that the OPPs were degraded rather than absorbed by the cells. Among the six selected strains, P9 was most tolerant to gastrointestinal juices and bile. We thus used ultra-high performance liquid chromatography electron spray ionization coupled with time-of-flight mass spectrometry (UPLC/ESI-Q-TOF/MS) to generate the metabolomic profiles of the strain P9 growing in MRS medium with and without containing phorate. By using orthogonal partial least squares discriminant analysis, we identified some potential phorate-derived degradative products. This work has identified novel lactic acid bacteria resources for application in pesticide degradation. Our results also shed light on the phorate degradation mechanism by P9.

Developing countries like Nigeria are increasingly employing pesticides to boost the productivity of their agriculture and food supply, despite the fact that doing so poses a health risk to the general populace. The purpose of this study was to assess pesticide residue levels in Lagos, Nigeria. A total of 18 samples from three neighbourhood markets were collected, and they were then examined for the presence of organochlorine (Endosulfan I, Pentachlorophenol, Heptachlor epoxide, and Endosulfan II) and organophosphate (Dichlorvos, Dimethoate, Phorate, and methyl parathion) residues. During the pre-treatment, the multi-residue Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS citrate) method with the addition of acetonitrile was used, and then samples were analysed using GC-MS. All of the samples contained dichlorvos, but the orange sample's concentration was below the limit of quantification, making quantification impossible. Dimethoate concentrations were below MRLs except waterleaf sample. With a concentration of 0.043 μg/mL, Waterleaf had the highest quantity of dimethoate in the sample. The findings of this study indicate that in order to safeguard the health of consumers, it is necessary to closely monitor organochlorine and organophosphate use nationwide, along with other related pesticides, and to test for pesticide residues in food products.

During two consecutive years of field trials containing 22 treatments, commercially prepared formulations of essential oils (EOs) were compared to an untreated control and to a standard chemical fumigant treatment (FU) (1,3-dichloropropene and metam sodium) applied preplant followed by phorate (PH) at planting to determine their value in the management of lesion nematode, and in improving plant health. The EO products Duogard, EF400, EF300, and Cinnamite were tested as preplant dips to bulblet planting stock. The treated bulblets were tested either alone, in combination with PH at-planting, at planting following FU or in combination with PH at planting following FU. The organophosphates ethoprop and fosthiazate were also tested either alone, or at a reduced rate in combination with a reduced rate of PH. With respect to bulb circumference, ten treatments consistently outperformed the control. In consecutive years, three treatments had healthier looking roots than the control. At harvest, levels of lesion nematode within roots were consistently lower in nine treatments. EOs were beneficial in mitigating nematode damage. In this work, a novel electrochemiluminescence (ECL) aptasensor was structured for the detection of four organophosphorus pesticides (OPs). Firstly, multi-walled carbon nanotubes (MWCNTs) were used to create a favorable loading interface for the fixation of tris (2, 2'-bipyridyl) ruthenium (II) (Ru (bpy)). At the same time, copper (core)-gold (shell) bimetallic nanoparticles (Cu@Au NPs) were synthesized in the aqueous phase for the sensor construction. Gold nanoparticles (Au NPs) could promote the electrochemiluminescence intensity of Ru (bpy) with high efficiency by catalyzing the oxidation process of tri-n-propylamine (TPrA). Compared with the Au NPs, Cu@Au NPs increased the solid loading of Au NPs by virtue of the large specific surface area of copper nanoparticles (Cu NPs), which could further improve the sensitivity of aptasensor. When OPs were added, the ECL intensity was significantly reduced, and the concentration of OPs could be detected through the ECL intensity. Under the optimum conditions, the aptasensor had a wider dynamic range and ultra-low detection limit for the detection of four pesticides: profenofos, isocarbophos, phorate, and omethoate, and their detection limits were 3 × 10 ng/mL, 3 × 10 ng/mL, 3 × 10 ng/mL, and 3 × 10 ng/mL respectively (S/N = 3). The aptasensor had the merits of good stability, reproducibility, and specificity, and had a favorable recovery rate in detecting OPs residues in vegetables. This work provided an effective method for the construction of a simple, rapid, and sensitive biosensor.

BACKGROUND
Multiple-herbicide resistance in Lolium rigidum and other weed species is increasingly exerting pressure on herbicide discovery research for solutions against resistance-prone weeds. In this study we investigate: (i) the responses of L. rigidum populations and wheat to the new herbicide cinmethylin in comparison with other pre-emergence herbicides, (ii) the effect of seed burial depths on cinmethylin efficacy and crop selectivity, and (iii) the basis of cinmethylin selectivity in wheat.
RESULTS
Cinmethylin at 400 g ha controls herbicide-susceptible and multiple-resistant L. rigidum, with a reduction of >85% in plant emergence and 90% in aboveground biomass. Cinmethylin provides effective control of a large number of field populations of L. rigidum with evident resistance to trifluralin. When the wheat seed is buried ≥1 cm below the cinmethylin-treated soil surface, the emergence of crop seedlings is not different from the untreated control. The organophosphate insecticide phorate synergizes cinmethylin toxicity in wheat, with an LD of 682 g ha in the absence of phorate versus 109 g ha in the presence of phorate (84% reduction). The synergistic effect of phorate with cinmethylin on herbicide-susceptible L. rigidum appears smaller (a 44% reduction in the LD of cinmethylin).
CONCLUSIONS
Cinmethylin is effective in controlling multiple-resistant L. rigidum and appears safe for wheat when the seed is separated at depth from the herbicide applied to the soil surface. The basis of this metabolism-based selectivity is likely regulated by cytochrome P450 monooxygenases. © 2020 Society of Chemical Industry.

Organophosphate pesticides are widely used; however, their use is limited due to neurotoxicity and, to a lesser extent, cardiotoxicity in humans. Given the high energy demands of cardiac muscle, which is characterized by a dense population of mitochondria, any damage to these organelles can exacerbate cardiotoxicity. This study aims to elucidate whether the cardiotoxic effects of organophosphate pesticides originate from mitochondrial dysfunction. To investigate this, toxicogenomic analyses were performed using various tools, such as the Comparative Toxicogenomic Database, GeneMANIA, STRING, and Cytoscape. Results revealed that 11 out of the 13 WHO-recommended Class Ia organophosphate pesticides target genes associated with cardiotoxicity. Notably, three of these genes were mitochondrial, with catalase (CAT) being the common differentially expressed gene among parathion, methyl parathion, and phorate. Furthermore, protein-protein interaction analysis indicated a strong association between CAT and superoxide dismutase 2, mitochondrial (SOD2). Subsequently, isolated heart mitochondria were utilized to assess CAT and superoxide dismutase (SOD) activities . The findings demonstrated that at a concentration of 7.5 ng/µL, both methyl parathion and phorate significantly decreased CAT activity by approximately 35%. Moreover, phorate reduced total SOD and SOD2 activities by 17% and 19%, respectively, at the same concentration. In contrast, none of the three organophosphate pesticides induced the opening of the mitochondrial permeability transition pore. These results suggest that the reduction in CAT and SOD2 activities, critical antioxidant enzymes, leads to the accumulation of reactive oxygen species within mitochondria, ultimately resulting in mitochondrial damage. This mechanism likely underlies the observed cardiotoxicity induced by these organophosphate pesticides.

Wireworms are primary pests of potatoes in Canada. Presently, the highly toxic organophosphate phorate (i.e., Thimet 20G) is the only effective insecticide in use in Canada. As such, there is an urgent need for novel alternative treatments that provide competitive tuber blemish protection and wireworm reduction with a safer human and environmental portfolio. Herein we evaluated broflanilide, a novel meta-diamide insecticide for both tuber protection and wireworm mortality. When evaluated in field trials in Agassiz, British Columbia over 6 yr, broflanilide applied as a seed piece treatment (SPT) to mother tubers at 1.5-2.0 g AI/100 kg seed (approx. 50 g AI/ha), or as an in-furrow spray (IFS) at 0.23-0.25 g AI/100 m row (approx. 25 g AI/ha) was as effective at reducing blemishes to daughter tubers by wireworms (Agriotes obscurus) as phorate (Thimet 20G at 3230 g AI/ha), bifenthrin (Capture 2EC IFS at 300 g AI/ha) and clothianidin (Titan ST at 312.5 g AI/ha). In addition, broflanilide SPT and IFS applied at the above rates reduced resident wireworms (in the field at the time of planting) by 95.4-99.0% and neonate wireworms (produced from eggs laid during the growing season) by 98.1-100%. Similar results were obtained when broflanilide IFS (nonsystemic) was paired with clothianidin SPT (systemic) for broad-spectrum potato insect pest control. Strategies for the use of broflanilide on wheat (e.g., Teraxxa F4) in rotation with potatoes (Cimegra), both registered in Canada in 2020 are discussed.

Organothiophosphate pesticides (OPPs) are the most commonly used pesticides, and their environmental migration brings serious water pollution and significant danger to human health, and thus it is urgent to develop effective technologies for removal of OPPs from water. Herein, magnetic covalent organic framework (COF) with a triazine skeleton was fabricated for enhanced adsorption and removal of OPPs from water. Magnetic COF has a fluffy ball-like structure, high crystallinity, large BET surface area (1543 m g), and regular mesopores (~3.1 nm). Therefore, it displayed high adsorption rates and large adsorption capacities for four typical OPPs, pyridafenthion, phoxim, pyrimitate, and phorate. Based on adsorption kinetic and isotherms investigations, the batch experimental data of magnetic COF was effectively modeled by pseudo-second-order kinetics and the Freundlich isothermal model. The equilibrium adsorption capacities of magnetic COF composite for OPPs ranged from 163.9 to 178.6 mg g, which were about 10 times higher than the amorphous magnetic composite. The adsorption mechanism was further explored to verify the contributions of π-π, CH⋯π, and CH⋯S interactions to the adsorption of OPPs on the crystalline magnetic COF. Furthermore, the high removal rate of OPPs from the environmental water and reusability further indicated its potential in real applications as an effective adsorption material.

BACKGROUND
Rice is a major field crop of paramount importance for global food security. However, the increased adoption of more profitable and resource-efficient direct-seeded rice (DSR) systems has contributed to greater weed infestations, including weedy rice, which has become a severe problem in several Asian regions. In this study we have developed a conceptually novel method to protect rice plants at high doses of clomazone and triallate.
RESULTS
The insecticide phorate applied to rice seeds provided a substantial level of protection against the herbicides clomazone or triallate. A quantity of 15 kg phorate ha significantly increased the LD values, which were more than twofold greater than for rice plants treated only with clomazone. A quantity of 20 kg phorate ha in combination with 2000 g triallate ha safened rice plants (80% survival) with LD >3.4-fold greater than in phorate-untreated rice. Weed control efficacy was not lowered by the presence of phorate-treated rice seeds.
CONCLUSION
Weedy rice is one of the most damaging global weeds and a major threat to DSR systems. In this study we have developed a proof-of-concept method to allow selective weedy rice control in rice crops. We call for herbicide discovery programmes and research to identify candidate safener and herbicide combinations to achieve selective herbicide control of weedy rice and alleviate weed infestations in global rice crops. © 2016 Society of Chemical Industry.
