BACKGROUND AND AIMS
Persistent organic pollutants (POPs) have multiple adverse effects on human health. Recent studies show a possible association with liver disease, but population-based data are scarce. In this population-based study, we studied the associations between POPs and biomarkers of liver disease and incident liver disease.
METHODS
This study consisted of 2789 adults that participated in the environmental toxin subset of the Finnish health-examination survey, FINRISK 2007. Toxins were measured from serum samples, and standard liver tests and dynamic aspartate aminotransferase-alanine aminotransferase ratio (dAAR) were measured as biomarkers of liver function. Associations between POPs and the biomarkers were then analysed using linear regression. Associations between POPs and incident liver disease (n = 36) were analysed by Cox regression.
RESULTS
Organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs) and several perfluorinated alkyl substances exhibited statistically significant positive associations with several biomarkers of liver injury (betacoefficient per SD 0.04-0.14, p < 0.05). These associations were stronger in subgroups of individuals with obesity or non-alcoholic fatty liver disease. OCPs, PCBs and perfluoro-octanoic acid also had significant positive associations with dAAR, which can be used to predict risk of incident severe liver outcomes (beta coefficient per SD 0.05-0.08, p < 0.05). OCPs and PCBs were also significantly and positively associated with incident liver disease (hazard ratio per SD 1.82 95% CI 1.21-2.73, p < 0.01 and hazard ratio per SD 1.69, 95% CI 1.07-2.68, p < 0.05 respectively).
CONCLUSIONS
Several POPs show positive associations with markers of liver injury and incident liver disease, suggesting that environmental toxins are important risk factors for chronic liver disease.

As nanoplastics and persistent organic pollutants are broadly distributed in aquatic ecosystems and pose a potential threat to ecosystem, most pertinent studies have focused on aquatic animals, while studies on freshwater plants have been rarely reported. Therefore, we analyzed the single and combined toxicological impacts of various concentrations of 80 nm polystyrene nanoplastics (PS-NPs) including 0.5, 5, 10, and 20 mg/L and polychlorinated biphenyl-52 (PCB-52, 2,2',5,5'- tetrachlorobiphenyl) at 0.1 mg/L on the aquatic plant Spirodela polyrhiza (S. polyrhiza) after a 10-day hydroponic experiment. Laser confocal scanning microscopy (LCSM) showed the accumulation of PS-NPs mainly in the root surface and the lower epidermis of leaves, and the enrichment of PS-NPs was aggravated by the presence of PCB-52. PS-NPs at 10 mg/L and 20 mg/L alone or in combination with PCB-52 notably inhibited the growth of S. polyrhiza, reduced the synthesis of chlorophylls a and b, and increased the activities of superoxide dismutase (SOD) and peroxidase (POD) as well as malondialdehyde (MDA) levels, and induced osmotic imbalance (soluble protein and soluble sugar contents) (p < 0.05). However, a single treatment with low levels of PS-NPs had positive effects on the growth (0.5 mg/L) and photosynthetic systems (0.5, 5 mg/L) of S. polyrhiza, while co-exposure exacerbated the damaging impacts of PS-NPs on the antioxidant defense system of S. polyrhiza, which was more pronounced in the roots. Furthermore, correlation analysis revealed that plant growth parameters were positively correlated with chlorophyll a and b content and negatively correlated with soluble sugars, antioxidant enzymes, lipid peroxidation, and carotenoid content (p < 0.05). These results provide data to improve the understanding of the single and combined ecotoxicological effects of nanoplastics and polychlorinated biphenyls (PCBs) in aquatic plants and their application in phytoremediation measures.

Accumulating number of epidemiological studies has recently proposed that improvement in the risk of autism spectrum disorders (ASD) is associated with persistent organic pollutants (POPs) exposure. However, evidence from current researches is limited and inconsistent. Thus, we conducted a systematic review and meta-analysis to investigate the potential associations comprehensively. We systematically and extensively searched two electronic databases (PubMed and EMBASE) from inception to July 3, 2022 and an updated search was performed before submission. Summary odds ratios (ORs) and 95% confidence intervals (CIs) were derived from stratified random-effects meta-analyses by type of exposure and outcome. We also tested the potential heterogeneity across studies, conducted sensitivity analysis and evaluated publication bias. A total of 20 studies were finally included in our study. Meta-analytical effect estimates indicated a positive association between prenatal exposure to PCB-138, PCB-153 and PCB-170 and an increased risk of ASD, with OR of 1.89 (95% CI = 1.21-2.95, I = 0%), 1.61 (95% CI = 1.05-2.47, I = 0%) and 1.46 (95% CI = 1.03-2.06, I = 0%) respectively. In contrast, PFDA was found inversely associated with the risk of ASD (OR = 0.70, 95% CI = 0.52-0.94, I = 0%). The level of evidence supporting a link between ASD risk and exposure to PCB-138, PCB-153, PCB-170, and PFDA was respectively categorized as low, low, moderate, and low. In summary, this systematic review and meta-analysis suggest that exposure to PCB-138, PCB-153, and PCB-170 correlates with a heightened risk of ASD, with evidence levels rated as "low", "low", and "moderate", respectively. In contrast, PFDA exposure appears to be inversely associated with ASD risk, with a "low" level of supporting evidence. However, due to the limited number of studies available for each exposure and outcome pairing, these results should be interpreted with caution. Sufficiently powered studies are needed to validate our findings.

Understanding the environmental and human impacts associated with polychlorinated dibenzo-p-dioxins/dibenzofurans (PCDD/Fs) and dioxin-like polychlorinated biphenyls (DL-PCBs) exposure from municipal solid waste incinerators (MSWIs) is challenging because information on ambient and dietary exposure levels, spatial characteristics, and potential exposure routes is limited. In this study, 20 households from two villages located on the upwind and downwind sides of a MSWI were selected to characterize the concentration and spatial distribution of PCDD/F and DL-PCB compounds in ambient and food samples, such as dust, air, soil, chicken, egg, and rice samples. The source of exposure was identified using congener profiles and principal component analysis. Overall, the dust and rice samples had the highest and lowest mean dioxin concentrations, respectively. Significant differences were observed (p < 0.01) in PCDD/F concentrations in chicken samples and DL-PCB concentrations in rice and air samples between the upwind and downwind villages. The exposure assessment indicated that the primary risk source was dietary exposure, especially from eggs, which had a PCDD/F toxic equivalency (TEQ) range of 0.31-14.38 pg TEQ/kg body weight (bw)/day, leading to adults in one household and children in two households exceeding the World Health Organization-defined threshold of 4 pg TEQ/kg bw/day. Chicken was the main contributor to the differences between upwind and downwind exposure. Based on the established congener profiles, the exposure routes of PCDD/Fs and DL-PCBs from the environment to food to humans were clarified.

The plasma membrane lipid rafts are cholesterol- and sphingolipid-enriched domains that allow regularly distributed, sub-micro-sized structures englobing proteins to compartmentalize cellular processes. These membrane domains can be highly heterogeneous and dynamic, functioning as signal transduction platforms that amplify the local concentrations and signaling of individual components. Moreover, they participate in cell signaling routes that are known to be important targets of environmental toxicants affecting cell redox status and calcium homeostasis, immune regulation, and hormonal functions. In this work, the evidence that plasma membrane raft-like domains operate as hubs for toxicants' cellular actions is discussed, and suggestions for future research are provided. Several studies address the insertion of pesticides and other organic pollutants into membranes, their accumulation in lipid rafts, or lipid rafts' disruption by polychlorinated biphenyls (PCBs), benzo[a]pyrene (B[a]P), and even metals/metalloids. In hepatocytes, macrophages, or neurons, B[a]P, airborne particulate matter, and other toxicants caused rafts' protein and lipid remodeling, oxidative changes, or amyloidogenesis. Different studies investigated the role of the invaginated lipid rafts present in endothelial cells in mediating the vascular inflammatory effects of PCBs. Furthermore, in vitro and in vivo data strongly implicate raft-localized NADPH oxidases, the aryl hydrocarbon receptor, caveolin-1, and protein kinases in the toxic mechanisms of occupational and environmental chemicals.

The current study examined the level of Polychlorinated biphenyls (PCBs) in tumor and blood serum of female breast cancer patients and control individuals recruited from Punjab, Pakistan. Breast tumor and blood serum from 40 patients and only blood serum from ten control subjects were obtained and concentration of 32 PCB congeners was analyzed through Gas chromatography coupled with Mass spectrophotometry. Sociodemographic variables of the patients along with essential clinical and haematological parameters were taken as covariates. Tumor reflects the highest median (min-max) concentration (ng g lw) of ƩPCBs at 115.94 (0.05-17.75) followed by 16.53 (0.09-2.94) and 5.24 (0.01-0.59) in blood serum of cancer patients and control group respectively. Median concentrations (ng g lw) of non-dioxine like ƩPCBs were considerably higher at 83.04, 32.89 and 4.27 compared to 13.03 and 3.50 and 0.97 for dioxin like ƩPCBs in tumor, serum of breast cancer patients and control subjects respectively. PCB-87 was most dominant congeners in tumor followed by PCB-170 and -82 whereas PCB-28 and -52 reflected greatest contribution in serum of breast cancer patients. Blood haemoglobin, potassium and chloride ions showed significant positive whereas body mass index reflect inverse relationship when regressed with ƩPCBs in tumor. This pioneer study depicts elevated concentrations of PCBs in patients compared to control, reflecting potential positive association of PCBs with breast cancer which need further confirmation. We concluded that chronic exposure to PCBs might be associated with an increasing number of breast cancer incidences in developing countries like Pakistan, which should be further elucidated through detail in vitro and in vivo studies.

Compound specific isotope analysis was extensively used to characterise the environmental processes associated with the abiotic and biotic transformation of persistent halogenated organic pollutants including those of contaminants of emerging concern (CECs). In the last years, the compound specific isotope analysis was applied as tool to evaluate the environmental fate and was expanded to larger molecules like brominated flame retardants and polychlorinated biphenyls. Multi-element (C, H, Cl, Br) CSIA methods have been also employed both in laboratory and field experiments. Nevertheless, despite the instrumental advances of isotope ratio mass spectrometers systems, the instrumental detection limit for gas chromatography-combustion-isotope ratio mass spectrometer (GC-C-IRMS) systems is challenging, especially when it is utilized to δC analysis. Liquid chromatography-combustion isotope ratio mass spectrometry methods are challenging, taking into consideration the chromatographic resolution required when analysing complex mixtures. For chiral contaminants, enantioselective stable isotope analysis (ESIA) has turned up as alternative approach but, up to now, it has been used for a limited number of compounds. Taking into consideration the occurrence of new emerging halogenated organic contaminants, new GC and LC methods for non-target screening using high resolution mass spectrometry are needed to be developed prior to the compound specific isotope analysis (CSIA) methods.

Chlorinated organic compounds, such as polychlorinated biphenyls (PCBs), are a threat to both humans and the environment because of their toxicity, persistence, and capacity for long-range atmospheric transport. The concentrations of 28 PCB congeners, including 12 dioxin-like and seven indicator PCBs, were investigated in soils, and indoor and outdoor dusts from Port Harcourt city, Nigeria, in order to evaluate the characteristic distribution patterns in these media, their sources, and possible risk. The PCB concentrations varied from 4.59 to 116 ng g for soils, and from 1.80 to 23.0 ng g and 2.73 to 57.4 ng g for indoor and outdoor dusts respectively. The sequence of PCB concentrations in these matrices was soil > outdoor dust > indoor dust. The composition of PCBs in these matrices indicated the prevalence of lower chlorinated PCBs in indoor and outdoor dusts, while the higher chlorinated congeners were dominant in soils. Di-PCBs were the predominant homologues in indoor dusts, while deca-PCBs were the most prevalent homologues in outdoor dusts and soils. The TEQ values of dioxin-like PCBs in 60 % of the soils, 100 % of the indoor dust, and 30 % of the outdoor dust were above the indicative value of 4 pg TEQ g established by the Canadian authority. The hazard index (HI) values for exposure of adults and children to PCBs in these media were mostly greater than one, while the total cancer risk (TCR) values exceeded the acceptable risk value of 10, which indicate probable non-carcinogenic and carcinogenic risks resulting from exposure to PCBs in these media. Source analysis for PCBs in these matrices shows that they originated from diverse sources.

Polychlorinated biphenyls (PCBs) are typical persistent organic pollutants that have been associated with type 2 diabetes (T2DM) in cohort studies. This review aims to comprehensively assess the molecular mechanisms of PCBs-induced T2DM. Recent progress has been made in the research of PCBs in liver tissue, adipose tissue, and other tissues. By influencing the function of nuclear receptors, such as the aryl hydrocarbon receptor (AhR), pregnancy X receptor (PXR), and peroxisome proliferator activated receptor γ (PPARγ), as well as the inflammatory response, PCBs disrupt the balance of hepatic glucose and lipid metabolism. This is associated with insulin resistance (IR) in the target organ of insulin. Through androgen receptor (AR), estrogen receptor α/β (ERα/β), and pancreato-duodenal-homeobox gene-1 (PDX-1), PCBs affect the secretion of insulin and increase blood glucose. Thus, this review is a discussion on the relationship between PCBs exposure and the pathogenesis of T2DM. It is hoped to provide basic concepts for diabetes research and disease treatment.

Single-pollutant methods to evaluate associations between endocrine disrupting chemicals (EDCs) and thyroid cancer risk may not reflect realistic human exposures. Therefore, we evaluated associations between exposure to a mixture of 18 EDCs, including polychlorinated biphenyls (PCBs), brominated flame retardants, and organochlorine pesticides, and risk of papillary thyroid cancer (PTC), the most common thyroid cancer histological subtype. We conducted a nested case-control study among U.S. military servicemembers of 652 histologically-confirmed PTC cases diagnosed between 2000 and 2013 and 652 controls, matched on birth year, sex, race/ethnicity, military component (active duty/reserve), and serum sample timing. We estimated mixture odds ratios (OR), 95% confidence intervals (95% CI), and standard errors (SE) for associations between pre-diagnostic serum EDC mixture concentrations, overall PTC risk, and risk of histological subtypes of PTC (classical, follicular), adjusted for body mass index and military branch, using quantile g-computation. Additionally, we identified relative contributions of individual mixture components to PTC risk, represented by positive and negative weights (w). A one-quartile increase in the serum mixture concentration was associated with a non-statistically significant increase in overall PTC risk (OR = 1.19; 95% CI = 0.91, 1.56; SE = 0.14). Stratified by histological subtype and race (White, Black), a one-quartile increase in the mixture was associated with increased classical PTC risk among those of White race (OR = 1.59; 95% CI = 1.06, 2.40; SE = 0.21), but not of Black race (OR = 0.95; 95% CI = 0.34, 2.68; SE = 0.53). PCBs 180, 199, and 118 had the greatest positive weights driving this association among those of White race (w = 0.312, 0.255, and 0.119, respectively). Findings suggest that exposure to an EDC mixture may be associated with increased classical PTC risk. These findings warrant further investigation in other study populations to better understand PTC risk by histological subtype and race.
