In dental implantology, alveolar ridge preservation (ARP) has emerged as a standard technique to address dimensional changes that affect alveolar ridge morphology following tooth loss. Various alternative graft materials, including xenografts, alloplasts, and allografts, have been effectively employed in fresh extraction sites for ARP. Current evidence suggests that these materials primarily serve as bio-scaffolds, which are slowly incorporated, thus necessitating a waiting period of at least 4-6 months before implant placement. Consequently, the ARP technique extends the overall duration of implant treatment by several months. Recently, the incorporation of a form of autologous platelet concentrate, known as platelet-rich fibrin (PRF), has been advocated in conjunction with ARP as a method of bioenhancement of soft- and hard-tissue healing and regeneration. PRF contains platelet-derived growth factors, hormones, and bioactive components like cytokines that have demonstrated the ability to stimulate angiogenesis and tissue regeneration throughout all phases of wound healing. Additionally, the concentration of leukocytes present in the PRF matrix plays a vital role in tissue healing and regeneration as part of the osteoimmune response. The reported advantages of incorporating autogenous PRF platelet concentrates during ARP encompass reduced healing time, improved angiogenesis and bone regeneration, socket sealing through the fibrin matrix, antibacterial properties, and decreased post-extraction pain and infection risk. Therefore, the objective of this paper is to review the existing evidence regarding the application of PRF in alveolar ridge preservation (ARP) following tooth extraction. Two clinical case studies are presented, wherein ARP was enhanced with PRF, followed by implant placement within a relatively short period of 8 weeks. These cases serve as further proof of concept for supporting the adjuvant use of PRF to enhance healing and accelerate implant placement after ARP.

L. is a plant with a wide range of potential medicinal applications. In recent years, polyploidy has gained attention as a potential strategy for rapidly improving , which, unlike other modern crops, has not yet benefitted from this established biotechnological application. Currently, no reports on high THCA and CBDA drug-type polyploid cultivars have been published. Moreover, it still needs to be clarified if different cultivars react similarly to polyploidization. For these reasons, we set out to evaluate and compare the phenotype and chemotype of three high Δ-tetrahydrocannabinolic acid (THCA) and one high cannabidiolic acid (CBDA) drug-type cultivars in their diploid, triploid and tetraploid state through agronomic and metabolomic approaches. Our observations on plant morphology revealed a significant increase in plant height and leaf size with increasing ploidy levels in a cultivar-dependent manner. In contrast, cannabinoids were negatively affected by polyploidization, with the concentration of total cannabinoids, THCA, CBDA and cannabigerolic acid (CBGA) decreasing significantly in higher ploidy levels across all four cultivars. Headspace analysis of volatiles revealed that total volatile content decreased in triploids. On the other hand, tetraploids reacted differently depending on the cultivars. Two THCA dominant cultivars showed an increase in concentrations, while in the other two cultivars, concentrations decreased. Additionally, several rare compounds not present in diploids appeared in higher ploidy levels. Moreover, in one high THCA cultivar, a couple of elite tetraploid genotypes for cannabinoid and volatile production were identified, highlighting the role of cultivar and genotypic variability as an important factor in L. polyploids. Overall, our observations on plant morphology align with the giga phenotype observed in polyploids of other plant species. The decrease in cannabinoids and volatiles production in triploids have relevant implications regarding their commercial use. On the other hand, this study found that tetraploidization is a suitable approach to improve L. medicinal potential, although the response is cultivar and genotype-dependent. This work lays the ground for further improving, evaluating and harnessing L. chemical diversity by the breeding, biotechnological and pharmaceutical sectors.

The role of pro-inflammatory cytokines in the toxicity of fluoride to tumor cells was investigated by culturing Hepa1-6 cells in medium containing gradient concentrations of fluoride (0, 0.5, 1, 1.5, 2, 3, 4, and 5 mmol/L). The viability of Hepa1-6 cells was detected via MTT assay. Interleukin (IL)-2, IL-6, tumor necrosis factor (TNF)-α, and IL-1β levels in the supernatant were determined via an enzyme-linked immunosorbent assay, and the protein expression levels of these enzymes in Hepa1-6 cells were evaluated by immunofluorescence staining. Results showed that the viability of Hepa1-6 cells remarkably decreases after fluoride exposure, especially at concentration of 3, 4, and 5 mmol/L fluoride. Levels of IL-2, TNF-α, and IL-1β in the supernatant markedly decreased when cells were exposed to fluoride at concentrations of 1 mmol/L or higher. However, levels of TNF-α and IL-1β substantially increased and IL-2 showed no remarkable change when the fluoride concentration was 0.5 mmol/L. The content of IL-6 remarkably increased with increasing fluoride concentrations up to 2 mmol/L, and then markedly decreased at 3, 4, and 5 mmol/L fluoride; the decreasing trend of IL-6 content under high fluoride exposure is consistent with the decrease in Hepa1-6 cell viability observed at the same concentration. The protein expression levels of IL-2, IL-6, TNF-α, and IL-1β were in accordance with their contents in the supernatant. In summary, our study demonstrated that fluoride inhibits Hepa1-6 cell growth and results in disorders in the expression and secretion pro-inflammatory cytokines.

Reputed as a significant metabolic disorder, non-alcoholic fatty liver disease (NAFLD) is characterized by high-fat deposits in the liver and causes substantial economic challenges to any country's workforce. Previous studies have indicated that some lactic acid bacteria may effectively prevent or treat NAFLD. Overall, KLDS1.0901 protected against HFD-induced NAFLD by improving liver characteristics and modulating microbiota composition, and thus could be a candidate for improving NAFLD. This study aimed to assess the protective effects of KLDS1.0901 on a high-fat diet(HFD)-induced NAFLD. First, hepatic lipid profile and histological alterations were determined to study whether KLDS1.0901 could ameliorate NAFLD. Then, the intestinal permeability and gut barrier were explored. Finally, gut microbiota was analyzed to elucidate the mechanism from the insights of the gut-liver axis. The results showed that KLDS1.0901 administration significantly decreased body weight, Lee's index body, fat rate, and liver index. KLDS1.0901 administration significantly improved lipid profiles by decreasing the hepatic levels of total cholesterol (TC), triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C) and by increasing the high-density lipoprotein cholesterol (HDL-C) levels. A conspicuous decrease of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum was observed after KLDS1.0901 administration. Meanwhile, the H&E and Oil Red O-stained staining showed that KLDS1.0901 significantly reduced liver lipid accumulation of HFD-fed mice by decreasing the NAS score and lipid area per total area. Our results showed that KLDS1.0901 administration decreased the interleukin-6 (IL-6), interleukin-1β (IL-1β), and tumor necrosis factor-alpha (TNF-α) concentrations accompanied by the increase of interleukin-10 (IL-10). KLDS1.0901 administration could improve the intestinal barrier function by upregulating the mRNA levels of occludin, claudin-1, ZO-1, and Muc-2, which were coupled to the decreases of the concentration of LPS and D-lactic acid. Notably, KLDS1.0901 administration modulated the gut microbiota to a near-normal pattern. Hence, our results suggested that KLDS1.0901 can be used as a candidate to ameliorate NAFLD.

A total of 131 sows (Line 241; DNA, Columbus, NE) were used in a study to evaluate the effect of increasing soybean meal concentration in lactating sow diets on sow and litter performance. Sows were blocked by body weight (BW) within parity on day 112 of gestation and allotted to one of three treatments of increasing dietary soybean meal (25%, 30%, or 35% of the total diet). Diets were formulated to 1.05% standardized ileal digestible lysine (Lys) with L-Lys HCl decreasing as soybean meal increased. All other amino acids and nutrients were formulated to meet nutrient requirement recommendations. Diets were fed from day 112 of gestation until weaning (day 20 ± 2). Litters were cross-fostered up to 48 h after farrowing to equalize litter size. Increasing soybean meal concentration increased (linear, = 0.017) sow BW loss and tended to increase (quadratic, = 0.052) sow backfat loss from farrowing to weaning. Sow average daily feed intake (ADFI) from day 0 to 7 was similar ( > 0.10) across dietary treatments. However, from day 7 to 14, 14 to weaning, and overall, ADFI decreased (linear, = 0.01) as soybean meal concentration increased. Despite the linear response in ADFI, the greatest decrease was observed as soybean meal concentration increased from 30% to 35% of the diet. There was no evidence for difference ( > 0.10) in wean-to-estrus interval, litter size, litter weight, or litter weight gain between dietary treatments. Sow serum urea nitrogen concentrations taken on day 14 of lactation increased (linear, = 0.001) as soybean meal concentration increased. There was no difference ( > 0.05) for sow creatinine concentration, regardless of dietary treatment, suggesting that the increased urea nitrogen was a reflection of the increased dietary crude protein as opposed to increased protein catabolism. In summary, sow feed intake decreased and weight loss increased as soybean meal concentration of the diet increased, with the greatest decrease observed at 35% of the total diet. Although there were no differences in litter performance, it appeared that 35% soybean meal in the lactation diet negatively affected feed intake.

Water and wastewater-based epidemiology have emerged as alternative methods to monitor and predict the course of outbreaks in communities. The recovery of microbial fractions, including viruses, bacteria, and microeukaryotes from wastewater and environmental water samples is one of the challenging steps in these approaches. In this study, we focused on the recovery efficiency of sequential ultrafiltration and skimmed milk flocculation (SMF) methods using Armored RNA as a test virus, which is also used as a control by some other studies. Prefiltration with 0.45 µm and 0.2 µm membrane disc filters were applied to eliminate solid particles before ultrafiltration to prevent the clogging of ultrafiltration devices. Test samples, processed with the sequential ultrafiltration method, were centrifuged at two different speeds. An increased speed resulted in lower recovery and positivity rates of Armored RNA. On the other hand, SMF resulted in relatively consistent recovery and positivity rates of Armored RNA. Additional tests conducted with environmental water samples demonstrated the utility of SMF to concentrate other microbial fractions. The partitioning of viruses into solid particles might have an impact on the overall recovery rates, considering the prefiltration step applied before the ultrafiltration of wastewater samples. SMF with prefiltration performed better when applied to environmental water samples due to lower solid concentrations in the samples and thus lower partitioning rates to solids. In the present study, the idea of using a sequential ultrafiltration method arose from the necessity to decrease the final volume of the viral concentrates during the COVID-19 pandemic, when the supply of the commonly used ultrafiltration devices was limited, and there was a need for the development of alternative viral concentration methods.

The objective of this study was to investigate whether the CH mitigation potential of 3-nitrooxypropanol (3-NOP) in dairy cattle was affected by basal diet (BD) composition. The experiment involved 64 Holstein-Friesian dairy cows (146 ± 45 d in milk at the start of trial; mean ± SD) in 2 overlapping crossover trials, each consisting of 2 measurement periods. Cows were blocked according to parity, d in milk, and milk yield, and randomly allocated to 1 of 3 diets: a grass silage-based diet (GS) consisting of 30% concentrates and 70% grass silage (DM basis), a grass silage- and corn silage-mixed diet (GSCS) consisting of 30% concentrates, 42% grass silage, and 28% corn silage (DM basis), or a corn silage-based diet (CS) consisting of 30% concentrates, 14% grass silage, and 56% corn silage (DM basis). Two types of concentrates were formulated, viz. a concentrate for the GS diet and a concentrate for the CS diet, to meet the energy and protein requirements for maintenance and milk production. The concentrate for the GSCS diet consisted of a 50:50 mixture of both concentrates. Subsequently, the cows within each type of BD received 2 treatments in a crossover design: either 60 mg of 3-NOP/kg of DM (NOP60) and a placebo with 0 mg of 3-NOP/kg of DM (NOP0) in one crossover or 80 mg of 3-NOP/kg of DM (NOP80) and NOP0 in the other crossover. Diets were provided as total mixed ration in feed bins, which automatically recorded feed intake. Additional concentrate was fed in the GreenFeed system that was used to measure emissions of CH and H. The CS diets resulted in a reduced CH yield (g/kg DMI) and CH intensity (g/kg milk). Feeding 3-NOP resulted in a decreased DMI. Milk production and composition did not differ between NOP60 and NOP0, whereas milk yield and the yield of major components decreased for NOP80 compared with NOP0. Feed efficiency was not affected by feeding 3-NOP. Interactions between BD and supplementation of 3-NOP were observed for the production (g/d) and yield (g/kg DMI) of both CH and H, indicating that the mitigating effect of 3-NOP depended on the composition of the BD. Emissions of CH decreased upon 3-NOP supplementation for all BD, but the decrease in CH emissions was smaller for GS (-26.2% for NOP60 and -28.4% for NOP80 in CH yield) compared with both GSCS (-35.1% for NOP60 and -37.9% for NOP80 for CH yield) and CS (-34.8% for NOP60 and -41.6% for NOP80 for CH yield), with no difference between the latter 2 BD. Emissions of H increased upon 3-NOP supplementation for all BD, but the H yield (g/kg DMI) increased 3.16 and 3.30-fold, respectively, when NOP60 and NOP80 were supplemented to GS, and 4.70 and 4.96 fold, respectively, when NOP60 and NOP80 were supplemented to CS. In conclusion, 3-NOP can effectively decrease CH emissions in dairy cows across diets, but the level of CH mitigation is greater when supplemented in a corn silage-based diet compared with a grass silage-based diet.

Obesity is a significant public health concern, and finding safe and effective means for combating this condition is needed. This study investigates the safety and efficacy of supplementation of a blend of capsaicinoids on weight gain, fat mass, and blood chemistry in a high-fat diet (HFD) model of obesity in mice and on adipocyte differentiation and gene expression in 3T3-L1 preadipocytes. High-fat diet (HFD)-fed mice were treated with a proprietary capsaicinoid concentrate (Capsimax; OmniActive Health Technologies Ltd., India) and compared to orlistat (ORL) and normal chow-fed mice (NC). Mice fed a high-fat diet showed significantly lower weight gain upon Capsimax (CAP) administration than their HFD counterparts and similar to that observed with ORL animals. In addition, CAP decreased the high-fat diet-induced increases in adipose tissue and epididymal fat pad mass and hypertrophy after 52 days of treatment. Both the CAP and ORL groups had increased plasma concentrations of leptin. CAP extracts decreased triacylglycerol content in 3T3-L1 preadipocytes and decreased markers of adipogenesis including peroxisome proliferator-activated receptor (PPAR-ɣ) and fatty acid-binding protein 4 (FABP4). Expression of genes involved in lipogenesis such as stearoyl-CoA desaturase (SCD) and fatty acid synthase (FSN) was decreased by CAP in a dose-dependent manner. Thermogenic genes and markers of brown adipose tissue including uncoupling protein 1 (UCP1) and PR domain-containing 16 (Prdm16) were induced by CAP in the preadipocyte cells. These in vivo and in vitro data support that this proprietary capsaicinoid concentrate reduces weight gain and adiposity at least in part through decreasing lipogenesis and increasing thermogenesis.

A microfluidic method was developed to study the ion-specific effect on bubble coalescence in salt solutions. Compared with other reported methods, microfluidics provides a more direct and accurate means of measuring bubble coalescence in salt solutions. We analyzed the coalescence time and approach velocity between bubbles and used simulation to investigate the pressure evolution during the coalescence process. The coalescence time of the three salt solutions decreased initially and then increased as the concentration of the salt solution was increased. The concentration with the shortest coalescence time is considered as the transition concentration (TC) and exhibits ion-specific. At the TC, the change in coalescence time indicates a shift in the effect of salt on bubble coalescence from facilitation to initial inhibition. Meanwhile, it can be seen that the sodium halide solutions significantly inhibit the bubble coalescence and the inhibition capability follows the order NaCl > NaBr > NaI. The results of the approach velocity show that the coalescence time decreases with increasing approach velocity, as well as the approach velocity was strongly influenced by concentration. The approach velocity undergoes a significant change at the TC. Furthermore, simulations of bubble coalescence in the microchannel indicate that the vertical pressure gradient at the center point of the bubble pairs increases as bubbles approach, driving liquid film drainage until bubble coalescence. The pressure at the center of the bubble pair reaches the maximum when the bubbles have first coalesced. It was further revealed that the concentration of the salt solution has a significant impact on the maximum pressure, as evidenced by the observed trend of decreasing pressure values with increasing concentrations.

Fine particulate matter (PM2.5) and near-surface ozone (O3) are the main atmospheric pollutants in China. Long-term exposure to high ozone concentrations adversely affects human health. It is of great significance to systematically analyze the spatiotemporal evolution mechanism and health effects of ozone pollution. Based on the ozone data of 91 monitoring stations in the Central Plains Urban Agglomeration from 2017 to 2020, the research used Kriging method and spatial autocorrelation analysis to investigate the spatiotemporal variations of ozone concentration. Additionally, the study assessed the health effects of ozone on the population using the population exposure risk model and exposure-response relationship model. The results indicated that: (1) The number of premature deaths caused by ozone pollution in the warm season were 37,053 at 95% confidence interval (95% CI: 28,190-45,930) in 2017, 37,685 (95% CI: 28,669-46,713) in 2018, and 37,655 (95% CI: 28,647-46,676) in 2019. (2) The ozone concentration of the Central Plains urban agglomeration showed a decreasing trend throughout the year and during the warm season from 2017 to 2020, there are two peaks monthly, one is June, and the other is September. (3) In the warm season, the high-risk areas of population exposure to ozone in the Central Plains Urban Agglomeration were mainly concentrated in urban areas. In general, the population exposure risk of the south is lower than that of the north. The number of premature deaths attributed to ozone concentration during the warm season has decreased, but some southern cities such as Xinyang and Zhumadian have also seen an increase in premature deaths. China has achieved significant results in air pollution control, but in areas with high ozone concentrations and high population density, the health burden caused by air pollution remains heavy, and stricter air pollution control policies need to be implemented.
