We have reported that transformed fetal liver cells produced a variant alpha-fetoprotein of 65K that differed from the mature alpha-fetoprotein of 69K and 73K in the polypeptide backbone. In the present study, we demonstrated that the biosynthetic pathway of the variant alpha-fetoprotein differed from that of the mature alpha-fetoprotein. The 65K variant was synthesized first as a preprotein of 49.5K which was processed to a polypeptide of 59K in the presence of microsomal membranes. The latter was the precursor of the variant alpha-fetoprotein found in cells and medium of transformed fetal liver cells. The 65K alpha-fetoprotein was encoded by a mRNA of 16S while mature AFP was encoded by a mRNA of 20S.

Purified human alpha-fetoprotein (HAFP) from five patients with hepatoma, one with gastric carcinoma, one with an embryonal cell tumor, and from fetal liver has demonstrated immunosuppressive potencies in vitro which vary over three orders of magnitude. A reversible association of HAFP with the cell surface and a predominant effect on T cells are suggested. No evidence of complex formation between HAFP and mitogen has been found. The microheterogeneity of HAFP has been detailed with crossed immunoelectrophoresis and isoelectric focusing in polyacrylamide gels containing 8M urea, and the immunosuppressive potency of HAFP isolated from a given source can be correlated with the proportion of certain HAFP species contained in it.

Adriamycin (ADM)-coated silica microspheres as a label for the sensitive detection of a cancer biomarker alpha-fetoprotein (AFP) was reported. Silica microspheres (SiO2 MSs) were employed as the carrier for the immobilization of gold nanoparticles (Au NPs), secondary antibody (Ab2) and ADM (denote: ADM@Au NPs@SiO2 MS/Ab2) as labels. In the presence of AFP, the labels were captured on the surface of the Au NP-reduced graphene oxide (rGO) (Au NP-rGO) nanocomposites to form a sandwich structure vs. the specific recognition of antibody-antigen. In a pH 7.4 phosphate buffer solution, a well-defined peak of ADM at about -0.70 V (vs. SCE) was recorded via differential pulse voltammetry, the peak intensity of which was related to the concentration of AFP. Under optimal experimental conditions, the immunoassay exhibited a wide linear range (0.5 pg mL-1 to 75 ng mL-1) and low limit of detection (0.17 pg mL-1). Further, the immunoassay was evaluated for serum samples, which gave satisfactory results.

An analysis has been made of indications for amniocentesis in the Edinburgh area from 1979 to 1981. About 5% of all mothers underwent the procedure. Among 2137 amniocenteses, 37% were performed on mothers 35 years old or more, and 30% on patients with raised serum alpha-fetoprotein. The total number of amniocenteses and the categories have been stable for the past three years. As a result of amniocentesis 104 pregnancies were terminated, 66 of which (63%) followed a raised maternal serum alpha-fetoprotein indication, while only 10 (9.6%) were in mothers aged 35 or more. There were a further 12 terminations based on raised serum alpha-fetoprotein but where no amniocentesis had been thought necessary. Even when figures for anencephaly are excluded from the analysis, maternal serum alpha-fetoprotein screening was responsible for detecting 35 out of 63 (56%) abnormal fetuses. This constitutes a strong case for the continuation of alpha-fetoprotein screening programmes.

A case of alpha-fetoprotein (AFP)-producing renal cell carcinoma with multiple bone and lung metastases, but without liver involvement is reported. The stain specific to AFP (avidin-biotin complex) proved the presence of the AFP in the cytoplasms of some cells of the renal tumors. In addition, three other cases of AFF-producing renal cancer recently published in the literature in Japan are reviewed. These four cases indicate that mesoderm-originating malignant tumors such as renal cell carcinomas can produce AFP in some situations. So, AFP is probably more universal than believed, although it is generally a popular and useful tumor marker for hepatocellular carcinomas and yolk sac tumors. The knowledge of the presence of AFP-producing renal cell carcinomas will make a new contribution to the study of the oncogenesis of AFP-producing tumors.
