INTRODUCTION
Dairy industry growth faces challenges in China due to inadequate forage, leading to high-concentrate diets and potential rumen issues. Buffering agents, like sodium bicarbonate, play a crucial role in stabilizing rumen pH. Alkaline Mineral Complex (AMC), a liquid additive with a pH of 14, shows promise in supporting dairy cow health and mitigating heat stress through ionization.
METHODS
This experiment was aimed to study the effect of adding AMC to total mixed ration (TMR) on ruminal fermentation and bacterial composition. AMCat 1, 2, 4, and 8 mL/kg was added to the substrate (0.5 g TMR). Nutrient digestibility was measured after 48 h fermentation, and fermentation parameters and microbial composition were measured after 48 h fermentation.
RESULTS AND DISCUSSION
The results of the experiment indicated that: The different concentrations of AMC showed a significant impact on time taken for gas production to reach 1/2 of the total gas production (HT) parameters ( < 0.05). Linear pH increase occurs at 6 and 24 h with rising AMC concentration ( < 0.05), showing a quadratic trend at 12 h ( < 0.05). The optimal buffering effect on rumen acid-base balance was observed at a 2 mL/kg concentration of AMC. Microbial diversity analysis indicated that there was no significant change in α-diversity with different AMC concentrations ( > 0.05). The microbial level demonstrated no significant difference in species diversity of rumen fluid bacteria among the various AMC concentration treatment groups compared to the control group, further supporting that the advantage of adding AMC in stabilizing the rumen environment without altering the structure of the rumen microbiota. Besides, the addition of AMC significantly increased the concentrations of acetate, propionate, total fatty acids (TVFA), and NH3-N, suggesting that AMC contributed to enhancing the energy and nitrogen utilization efficiency in ruminants. Based on the above detection indicators, we recommend that the most favorable concentration is 2 mL/kg.

A new method was established for the simultaneous analysis of four homologous benzalkonium chlorides (dodecyldimethylbenzyl ammonium chloride, tetradecyldimethylbenzyl ammonium chloride, hexadecyldimethylbenzyl ammonium chloride, and octadecyldimethylbenzyl ammonium chloride) in compound chemical disinfectants using non-aqueous capillary electrophoresis (CE) based on a micellar electrokinetic chromatography mode with direct ultraviolet detection. The separation was performed on an uncoated fused quartz capillary with a total length of 60.2 cm and a diameter of 25 μm. The separation buffer consisted of a mixture of methanol/acetonitrile (60:40, v/v) containing 70 mmol/L sodium acetate, 60 mmol/L trifluoroacetic acid and 20 mmol/L sodium dodecyl sulfate. The sample buffer was a methanol solution containing only 2 mmol/L trifluoroacetic acid. The separation voltage was set at 8 kV with a working current of approximately 2.3 μA. The detection wavelength was 214 nm. Under optimal conditions, the limit of detection and limit of quantification for these four benzalkonium chlorides (BACs) were 1.0 mg/L and 5.0 mg/L, respectively. Good linearities were observed in the concentration ranges from 5.0 to 100.0 mg/L, with correlation coefficients above 0.999 for all compounds. The recoveries of these four BACs ranged from 92.5 % to 109.1 % with relative standard deviations below 4.7 %. With the new method, all four BACs could be analyzed in a single injection. In contrast, the aqueous CE method in the National Standard GB/T 26369-2020 only allowed for the simultaneous analysis of the first three homologous. The new method demonstrated the improved peak shape compared to the aqueous CE method and then was successfully applied to the analysis of 19 commercially available samples, such as object table disinfectants, hand sanitizers, and disinfectant wipes, which claimed to contain quaternary ammonium compound. The results obtained using the new method were compared with those of the aqueous CE of the National Standard Method, and no statistically significant differences were observed. The new method is simple in pre-treatment and provides accurate results, making it highly suitable for routine analysis.

Clinical and Experimental Hypertension... Dec 2024Fibroblast growth factor 21 (FGF21) has a protective effect against cardiovascular disease. However, the role of FGF21 in hypertension remains elusive.
BACKGROUND
Fibroblast growth factor 21 (FGF21) has a protective effect against cardiovascular disease. However, the role of FGF21 in hypertension remains elusive.
METHODS
Ten-week-old male C57BL/6 mice were randomly divided into normal-salt (NS) group, NS+FGF21 group, deoxycorticosterone acetate-salt (DOCA) group and DOCA+FGF21 group. The mice in NS group underwent uninephrectomy without receiving DOCA and 1% NaCl and the mice in DOCA group were subjected to uninephrectomy and DOCA-salt (DOCA and 1% NaCl) treatment for 6 weeks. At the same time, the mice were infused with vehicle (artificial cerebrospinal fluid, aCSF) or FGF21 (1 mg/kg) into the bilateral paraventricular nucleus (PVN) of mice.
RESULTS
Here, we showed that FGF21 treatment lowered DOCA salt-induced inflammation and oxidative stress in the PVN, which reduced sympathetic nerve activity and hypertension. Mechanistically, FGF21 treatment decreased the expression of HNF4α and inhibited the binding activity of HNF4α to the promoter region of ACE2 in the PVN of DOCA salt-treated mice, which further up-regulated ACE2/Ang (1-7) signals in the PVN. In addition, ACE2 deficiency abolished the protective effect of FGF21 in DOCA salt-treated mice, suggesting that FGF21-mediated antihypertensive effect was dependent on ACE2.
CONCLUSIONS
The results demonstrate that FGF21 protects against salt-sensitive hypertension via regulating HNF4α/ACE2/Ang (1-7) axis in the PVN of DOCA salt-treated mice via multi-organ crosstalk between liver, brain and blood vessels.

Adsorption is an effective approach for remediating ammonium pollution, and zeolite has exceptional efficacy for the adsorption of ammonium. The investigation of ammonium adsorption using coal-fly-ash-based zeolite has gained remarkable attention in contemporary research. In this work, a sodium-acetate-modified synthetic zeolite (MSZ) was used to absorb ammonium in simulated wastewater. The MSZ had an adsorption capacity for ammonium of 27.46 mg g, and the adsorption process followed the Langmuir isotherm model and pseudo-second-order kinetics model. The adsorption and desorption of ammonium were controlled by ion exchange, pore diffusion, and electrostatic attraction processes. Ion exchange was responsible for 77.90% of the adsorption process and 80.16% of the desorption process. The MSZ was capable of continuously removing large amounts of ammonium from wastewater through fixed bed adsorption. After 5 regeneration cycles, MSZ still maintained 75% adsorption characteristics for ammonium. Using MSZ adsorbed with ammonium as a soil amendment increased the germination rate of mung beans by 10%. Furthermore, it also increased the stem length, root length, and fresh weight by 20-30%. These findings suggest that MSZ provides a promising application prospect to mitigate ammonium pollution and recycle ammonium resources.

Completion fluids play a vital role in well-related processes within the oil extraction industry. This article presents a comprehensive study of the properties and performance of various brine solutions as completion fluids for different well and reservoir conditions. Attributes examined include density, corrosion resistance, temperature stability, compatibility with formation fluids, clay swelling potential and influence on wettability. The research highlights the significance of selecting appropriate completion fluids to optimize well and reservoir operations. Zinc chloride emerges as an excellent option for high density applications, while sodium chloride and potassium formate solutions are ideal for extreme cold conditions. Potassium acetate outperforms calcium chloride and potassium chloride and has excellent pH stability. The compatibility of completion fluids with formation water has been observed to be excellent, with no sedimentation or emulsion formation. Potassium acetate also experiences minimal clay swelling, making it suitable for clay-rich formations. On the other hand, calcium chloride has a higher clay swelling than most of the brines tested, making it less suitable for sandstone formations with a higher clay content than these brines. The research evaluates the water-wetting abilities of completion fluids in carbonate and sandstone formations. Potassium chloride and zinc chloride have the most significant impact in carbonate formations, while potassium acetate and potassium formate excel in sandstone formations. This study provides a comprehensive understanding of completion fluids, facilitating informed decisions that maximize operational efficiency, protect reservoir integrity, and enhance hydrocarbon recovery. The appropriate selection of completion fluids should align with specific well and reservoir conditions, considering the priorities of the application.

In this work, we propose the formation of stretchable hydrogels at neutral pH from the physical crosslinking of chitosan (CS) and hyaluronic acid (HA) by polyelectrolyte complexation. A mixture of CS (M ≈ 600 kg/mol, degree of acetylation ≈ 50 %) solution and HA (M ≈ 77 kg/mol) solution was prepared with an excess of salts screening the electrostatic interactions CS/HA. In a controlled manner, the polyelectrolyte complexation was induced through the progressive dialysis of the salted polymer mixture against a sodium acetate solution (AcONa, 0.01 M) for 7 days. Depending on [HA], various materials were obtained: viscous solutions at [HA] = 0.75 % (w/v); hydrogels at [HA] = 1.50-2.24 % (w/v) with Young modulus of 14 kPa and stretchable to 200 %. The small angle X-ray scattering characterization of the hydrogels revealed a multiscale organization related to the conformation of the polymers induced by the physical interactions. The dialysis process with AcONa was optimized by adding a dialysis step against a zinc acetate solution containing Zn. The combination of polyelectrolyte complexation between CS/HA and metal complexation between Zn and the polymers led to an enhancement of the hydrogel stretchability up to 400 %.

BACKGROUND
Although the pathogenesis of inflammatory bowel disease (IBD), including ulcerative colitis (UC) and Crohn's disease (CD), has not been fully elucidated, accumulating researches suggest that intestinal microbiota imbalance contributes to the development of IBD in patients and animal models. RDP58, a peptide-based computer-assisted rational design, has been demonstrated to be effective in protecting against a wide range of autoimmune and inflammatory diseases. However, the underlying mechanism by which RDP58 protects against IBD mediated by intestinal microbiota has yet to be elucidated.
METHODS
The colitis model was induced by continuously administering 2.5 % (wt/vol) dextran sodium sulfate (DSS) solution for 7 days. The manifestations of colon inflammation were assessed via daily weight changes, colon length, tumor necrosis factor-alpha (TNF-α) level, disease activity index (DAI) score, pathology score, and intestinal barrier permeability. Intestinal microbiota analysis was carried out by 16S-rRNA sequencing. Colonic short chain fatty acids (SCFAs) and regulatory T cells (Tregs) were also detected. To further confirm the protective effect of RDP58 on intestinal microbiota, broad-spectrum antibiotic cocktail (ABX) treatment and fecal microbial transplantation (FMT) experiment were performed.
RESULTS
Oral administration of RDP58 ameliorated DSS-induced mice colitis by altering the diversity and composition of intestinal microbiota. Notably, RDP58 significantly upregulated SCFAs-producing microbiota, thereby promoting the generation of Tregs. ABX and FMT were performed to verify the above mechanism.
CONCLUSIONS
RDP58 ameliorated DSS-induced colitis through altering intestinal microbiota and enhancing SCFAs and Tregs production in intestinal microbiota dependent manner, potentially provide a novel therapy for IBD.

BACKGROUND
Ulcerative colitis (UC) and systemic lupus erythematosus (SLE) are both systemic immunoreactive diseases, and their pathogenesis depends on the interaction between genes and environmental factors. There are no reports of UC with SLE in China, but six cases of SLE with UC have been reported in China. The combination of these two diseases has distinct effects on the pathogenesis of both diseases.
CASE SUMMARY
A female patient (30 years old) came to our hospital due to dull umbilical pain, diarrhea and mucous bloody stool in August 2018 and was diagnosed with UC. The symptoms were relieved after oral administration of mesalazine (1 g po tid) or folic acid (5 mg po qd), and the patient were fed a control diet. On June 24, 2019, the patient was admitted for treatment due to anemia and tinnitus. During hospitalization, the patient had repeated low-grade fever and a progressively decreased Hb level. Blood tests revealed positive antinuclear antibody test, positive anti-dsDNA antibody, 0.24 g/L C3 (0.9-1.8 g/L), 0.04 g/L C4 (0.1-0.4 g/L), 32.37 g/L immunoglobulin (8-17 g/L), and 31568.1 mg/24 h total 24-h urine protein (0-150 mg/24 h). The patient was diagnosed with SLE involving the joints, kidneys and blood system. Previously reported cases of SLE were retrieved from PubMed to characterize clinicopathological features and identify prognostic factors for SLE.
CONCLUSION
The patient was discharged in remission after a series of treatments, such as intravenous methylprednisolone sodium succinate, intravenous human immunoglobulin, cyclophosphamide injection, and plasma exchange. After discharge, the patient took oral prednisone acetate tablets, cyclosporine capsules, hydroxychloroquine sulfate tablets and other treatments for symptoms and was followed up regularly for 1 month, after which the patient's condition continued to improve and stabilize.

Comparative Study
Extracellular vesicles (EVs) are nano-sized particles involved in intercellular communications that intrinsically possess many attributes as a modern drug delivery platform. -derived EVs (HpEVs) can be potentially exploited as a high-value-added bioproduct during astaxanthin production. The encapsulation of HpEV cargo is a crucial key for the determination of their biological functions and therapeutic potentials. However, little is known about the composition of HpEVs, limiting insights into their biological properties and application characteristics. This study examined the protein composition of HpEVs from three growth phases of grown under high light (350 µmol·m·s) and sodium acetate (45 mM) stresses. A total of 2038 proteins were identified, the majority of which were associated with biological processes including signal transduction, cell proliferation, cell metabolism, and the cell response to stress. Comparative analysis indicated that cells sort variant proteins into HpEVs at different physiological states. It was revealed that HpEVs from the early growth stage of contain more proteins associated with cellular functions involved in primary metabolite, cell division, and cellular energy metabolism, while HpEVs from the late growth stage of were enriched in proteins involved in cell wall synthesis and secondary metabolism. This is the first study to report and compare the protein composition of HpEVs from different growth stages of , providing important information on the development and production of functional microalgal-derived EVs.

The normal growth and development of skeletal muscle is essential for the health of the body. The regulation of skeletal muscle by intestinal microorganisms and their metabolites has been continuously demonstrated. Acetate is the predominant short-chain fatty acids synthesized by gut microbiota through the fermentation of dietary fiber; however, the underlying molecular mechanisms governing the interaction between acetate and skeletal muscle during the rapid growth stage remains to be further elucidated. Herein, specific pathogen-free (SPF) mice, germ-free (GF) mice, and germ-free mice supplemented with sodium acetate (GS) were used to evaluate the effects of acetate on the skeletal muscle growth and development of young mice with gut microbiota deficiency. We found that the concentration of serum acetate, body mass gain, succinate dehydrogenase activity, and expression of the myogenesis maker gene of skeletal muscle in the GS group were higher than those in the GF group, following sodium acetate supplementation. Furthermore, the transcriptome analysis revealed that acetate activated the biological processes that regulate skeletal muscle growth and development in the GF group, which are otherwise inhibited due to a gut microbiota deficiency. The in vitro experiment showed that acetate up-regulated Gm16062 to promote skeletal muscle cell differentiation. Overall, our findings proved that acetate promotes skeletal muscle growth and development in young mice via increasing Gm16062 expression.
