Two recent seminal works have untangled the intricate role of tumor-associated senescent cells in cancer progression, or regression, by guiding our immune system against cancer cells. The characterization of these unique, yet diverse cell populations, should be considered, particularly when contemplating the use of senolytics, which are drugs that selectively eliminate senescent cells, in a cancer framework. Here, we will describe the current knowledge in this field. In particular, we will discuss how the presence of senescent cells in tumors could be used as a therapeutic target in immunogenic cancers and how we may hypothetically design an adaptive anti-aging vaccine.

To humans, the diverse array of display behaviors that animals use for communication can easily seem peculiar or bizarre. While ample research delves into the evolutionary principles that shape these signals' effectiveness, little attention is paid to evolutionary patterning of signal design across taxa, particularly when it comes to the potential convergent evolution of many elaborate behavioral displays. By taking a mechanistic perspective, we explore the physiological and neurobiological mechanisms that likely influence the evolution of communication signals, emphasizing the utilization of pre-existing structures over novel adaptations. Central to this investigation are the concepts of perceptual bias and ritualization, which we propose contribute to the convergence of elaborate display designs across species. Perceptual bias explains a phenomenon where pre-existing perceptual systems of receivers, used for innate behaviors such as food and predator recognition, select for certain traits of a communication signal from a signaler. Ritualization occurs when traits with no functional role in communication are co-opted through selection and transformed into a new communicative signal. Importantly, susceptibility for ritualization can be brought about through physiological modifications that occurred early in evolutionary time. In this way, perceptual bias can be a selective force that causes the co-option of non-communicative traits into a new communication signal through ritualization involving pre-existing modifications to physiological systems. If the perceptual bias, non-communicative signal, and physiological modifications that increase susceptibility to ritualization are highly conserved, then we may see the convergent evolution of the new communication signal with unrelated taxa facing similar sensory constraints. We explore this idea here using the foot-flagging frog system as a theoretical case study.

Unraveling the mechanisms and responses of aniline-degrading biosystem to salinity stress in high temperature condition: pollutants removal performance and microbial community.
To explore the intrinsic influence of different salinity content on aniline biodegradation system in high temperature condition of 35±1 °C, six groups at various salinity concentration (0.0%-5.0%) were applied. The results showed that the salinity exerted insignificant impact on aniline removal performance. The low-level salinity (0.5%-1.5%) stimulated the nitrogen metabolism performance. The G5-2.5% had excellent adaptability to salinity while the nitrogen removal capacity of G6-5.0% was almost lost. Moreover, high throughput sequencing analysis revealed that the g__norank_f__NS9_marine_group, g__Thauera and g__unclassified_f__Rhodobacteraceae proliferated wildly and established positive correlation each other in low salinity systems. The g__SM1A02 occupying the dominant position in G5 ensured the nitrification performance. In contrast, the Rhodococcus possessing great survival advantage in tremendous osmotic pressure competed with most functional genus, triggering the collapse of nitrogen metabolism capacity in G6. This work provided valuable guidance for the aniline wastewater treatment under salinity stress in high temperature condition.

Characteristics such as low contrast and significant organ shape variations are often exhibited in medical images. The improvement of segmentation performance in medical imaging is limited by the generally insufficient adaptive capabilities of existing attention mechanisms. An efficient Channel Prior Convolutional Attention (CPCA) method is proposed in this paper, supporting the dynamic distribution of attention weights in both channel and spatial dimensions. Spatial relationships are effectively extracted while preserving the channel prior by employing a multi-scale depth-wise convolutional module. The ability to focus on informative channels and important regions is possessed by CPCA. A segmentation network called CPCANet for medical image segmentation is proposed based on CPCA. CPCANet is validated on two publicly available datasets. Improved segmentation performance is achieved by CPCANet while requiring fewer computational resources through comparisons with state-of-the-art algorithms. Spatial epigenetic mapping of tissues enables the study of gene regulation programs and cellular functions with the dependency on their local tissue environment. Here we outline a complete procedure for two spatial epigenomic profiling methods: spatially resolved genome-wide profiling of histone modifications using in situ cleavage under targets and tagmentation (CUT&Tag) chemistry (spatial-CUT&Tag) and transposase-accessible chromatin sequencing (spatial-ATAC-sequencing) for chromatin accessibility. Both assays utilize in-tissue Tn5 transposition to recognize genomic DNA loci followed by microfluidic deterministic barcoding to incorporate spatial address codes. Furthermore, these two methods do not necessitate prior knowledge of the transcription or epigenetic markers for a given tissue or cell type but permit genome-wide unbiased profiling pixel-by-pixel at the 10 μm pixel size level and single-base resolution. To support the widespread adaptation of these methods, details are provided in five general steps: (1) sample preparation; (2) Tn5 transposition in spatial-ATAC-sequencing or antibody-controlled pA-Tn5 tagmentation in CUT&Tag; (3) library preparation; (4) next-generation sequencing; and (5) data analysis using our customed pipelines available at: https://github.com/dyxmvp/Spatial_ATAC-seq and https://github.com/dyxmvp/spatial-CUT-Tag . The whole procedure can be completed on four samples in 2-3 days. Familiarity with basic molecular biology and bioinformatics skills with access to a high-performance computing environment are required. A rudimentary understanding of pathology and specimen sectioning, as well as deterministic barcoding in tissue-specific skills (e.g., design of a multiparameter barcode panel and creation of microfluidic devices), are also advantageous. In this protocol, we mainly focus on spatial profiling of tissue region-specific epigenetic landscapes in mouse embryos and mouse brains using spatial-ATAC-sequencing and spatial-CUT&Tag, but these methods can be used for other species with no need for species-specific probe design.

Metacognitive biases have been repeatedly associated with transdiagnostic psychiatric dimensions of 'anxious-depression' and 'compulsivity and intrusive thought', cross-sectionally. To progress our understanding of the underlying neurocognitive mechanisms, new methods are required to measure metacognition remotely, within individuals over time. We developed a gamified smartphone task designed to measure visuo-perceptual metacognitive (confidence) bias and investigated its psychometric properties across two studies (N = 3410 unpaid citizen scientists, N = 52 paid participants). We assessed convergent validity, split-half and test-retest reliability, and identified the minimum number of trials required to capture its clinical correlates. Convergent validity of metacognitive bias was moderate (r(50) = 0.64, p < 0.001) and it demonstrated excellent split-half reliability (r(50) = 0.91, p < 0.001). Anxious-depression was associated with decreased confidence (β = - 0.23, SE = 0.02, p < 0.001), while compulsivity and intrusive thought was associated with greater confidence (β = 0.07, SE = 0.02, p < 0.001). The associations between metacognitive biases and transdiagnostic psychiatry dimensions are evident in as few as 40 trials. Metacognitive biases in decision-making are stable within and across sessions, exhibiting very high test-retest reliability for the 100-trial (ICC = 0.86, N = 110) and 40-trial (ICC = 0.86, N = 120) versions of Meta Mind. Hybrid 'self-report cognition' tasks may be one way to bridge the recently discussed reliability gap in computational psychiatry.

OBJECTIVES
Perioperative mental health of older Black surgical patients is associated with poor surgical outcomes; however, evidence-based perioperative interventions are lacking. Our two study objectives included: first, examine factors affecting perioperative care experiences of older Black surgical patients with mental health problems, and second, ascertain design and implementation requirements for a culturally-adapted perioperative mental health intervention.
DESIGN SETTING AND PARTICIPANTS
We conducted six focus groups with older Black patients (n = 15; ≥50 years; surgery within the past 5 years and/or interest in mental health research; history of distress, anxiety, or depression coping with surgery/hospitalization/) from a large academic medical center. We engaged study partners, including interventionists and community members, to gather insights on intervention and implementation needs. We followed a hybrid inductive-deductive thematic approach using open coding and the National Institute on Minority Health and Health Disparities Research Framework.
RESULTS
Patients reported that their psychological well-being and long-term mental health outcomes were not appropriately considered during perioperative care. Perceived stressors included interpersonal and structural barriers to using mental healthcare services, clinician treatment biases and ageism in care, and lack of healthcare professional connections/resources. Patients utilized various coping strategies, including talk therapy, faith/spirituality, and family and friends.
CONCLUSION
This study offers valuable insights into the experiences of older Black surgical patients and the critical elements for developing a personalized perioperative mental health intervention to support their well-being before, during, and after surgery. Our findings demonstrated a need for a patient-centered and culturally adapted intervention targeting the individual/behavioral and interpersonal levels. Informed by the cultural adaptation framework, we propose a multi-component intervention that integrates psychological and pharmacological components.

OBJECTIVE
To enhance the quality of low-resolution (LR) ultrasound images and mitigate artifacts and speckle noise, which can impede accurate medical diagnosis, a novel method called the dual frequency-domain guided adaptation model (DF-GAM) is proposed. The method aims to achieve high-quality image reconstruction across diverse domains, including different ultrasound machines, diseases and phantom images.
METHODS
DF-GAM utilizes a dual-branch network architecture combined with frequency-domain self-adaptation and self-supervised edge regression. This approach enables cross-domain enhancement by focusing on the reconstruction of clear tissue structures and speckle patterns. The model is designed to adapt to various ultrasound imaging (USI) scenarios, ensuring its applicability in real-world clinical settings.
RESULTS
Experimental evaluations of DF-GAM were conducted using five different datasets. The results demonstrated the method's effectiveness, with DF-GAM outperforming existing enhancement techniques. The average peak signal-to-noise ratio (PSNR) achieved was 34.62, and the structural similarity index (SSIM) was 0.91, indicating a significant improvement in image quality compared to other methods.
CONCLUSION
DF-GAM shows great potential in improving medical image diagnosis and interpretation. Its ability to enhance LR ultrasound images across various domains without the need for extensive training data makes it a valuable tool for clinical use. The high PSNR and SSIM scores validate the method's effectiveness, suggesting that DF-GAM could significantly contribute to the field of USI diagnostics.

OBJECTIVE
This study began as a single-blind randomized controlled trial (RCT) to investigate the efficacy and safety of electroconvulsive therapy (ECT) for severe treatment-refractory agitation in advanced dementia. The aims are to assess agitation reduction using the Cohen-Mansfield Agitation Inventory (CMAI), evaluate tolerability and safety outcomes, and explore the long-term stability of agitation reduction and global functioning. Due to challenges encountered during implementation, including recruitment obstacles and operational difficulties, the study design was modified to an open-label format and other protocol amendments were implemented.
METHODS
Initially, the RCT randomized participants 1:1 to either ECT plus usual care or simulated ECT plus usual care (S-ECT) groups. As patients were enrolled, data were collected from both ECT and simulated ECT (S-ECT) patients. The study now continues in an open-label study design where all patients receive actual ECT, reducing the targeted sample size from 200 to 50 participants.
RESULTS
Study is ongoing and open to enrollment.
CONCLUSION
The transition of the ECT-AD study design from an RCT to open-label design exemplifies adaptive research methodologies in response to real-world challenges. Data from both the RCT and open-label phases of the study will provide a unique perspective on the role of ECT in managing severe treatment-refractory agitation in dementia, potentially influencing future clinical practices and research approaches.

Identifying critical growth stage and resilient genotypes in cowpea under drought stress contributes to enhancing crop tolerance for improvement and adaptation in Cameroon.
Drought stress following climate change is likely a scenario that will have to face crop growers in tropical regions. In mitigating this constraint, the best option should be the selection and use of resilient varieties that can withstand drought threats. Therefore, a pot experiment was conducted under greenhouse conditions at the Research and Teaching Farm of the Faculty of Agronomy and Agricultural Sciences of the University of Dschang. The objectives are to identify sensitive growth stage, to identify drought-tolerant genotypes with the help of yield-based selection indices and to identify suitable selection indices that are associated with yield under non-stress and stress circumstances. Eighty-eight cowpea genotypes from the sahelian and western regions of Cameroon were subjected to drought stress at vegetative (VDS) and flowering (FDS) stages by withholding water for 28 days, using a split plot design with two factors and three replications. Seed yields under stress (Ys) and non-stress (Yp) conditions were recorded. Fifteen drought indices were calculated for the two drought stress levels against the yield from non-stress plants. Drought Intensity Index (DII) under VDS and FDS were 0.71 and 0.84 respectively, indicating severe drought stress for both stages. However, flowering stage was significantly more sensitive to drought stress compared to vegetative stage. Based on PCA and correlation analysis, Stress Tolerance Index (STI), Relative Efficiency Index (REI), Geometric Mean Productivity (GMP), Mean Productivity (MP), Yield Index (YI) and Harmonic Mean (HM) correlated strongly with yield under stress and non-stress conditions and are therefore suitable to discriminate high-yielding and tolerant genotypes under both stress and non-stress conditions. Either under VDS and FDS, CP-016 exhibited an outstanding performance under drought stress and was revealed as the most drought tolerant genotype as shown by ranking, PCA and cluster analysis. Taking into account all indices, the top five genotypes namely CP-016, CP-021, MTA-22, CP-056 and CP-060 were identified as the most drought-tolerant genotypes under VDS. For stress activated at flowering stage (FDS), CP-016, CP-056, CP-021, CP-028 and MTA-22 were the top five most drought-tolerant genotypes. Several genotypes with insignificant Ys and irrelevant rank among which CP-037, NDT-001, CP-036, CP-034, NDT-002, CP-031, NDT-011 were identified as highly drought sensitive with low yield stability. This study identified the most sensitive stage and drought tolerant genotypes that are proposed for genetic improvement of cowpea.
