Review
Reliance on modeling and simulation in drug discovery and development has dramatically increased over the past decade. Two disciplines at the forefront of this activity, pharmacometrics and systems pharmacology (SP), emerged independently from different fields; consequently, a perception exists that only few examples integrate these approaches. Herein, we review the state of pharmacometrics and SP integration and describe benefits of combining these approaches in a model-informed drug discovery and development framework.

It is fitting that the 100th anniversary of the Handbook of Experimental Pharmacology celebrates not only its founding but also the founding of experimental pharmacology as both had their beginnings in Germany. Founded in 1919 by Arthur Heffter (1859-1925) as the "Handbuch der Experimentellen Pharmakologie" and renamed to its current title in 1937, the Handbook has continued to capture the emergence and developments of experimental pharmacology since the initial systematic work of Rudolf Buchheim and his student Oswald Schmiedeberg. Heffter, the first Chairman of the German Society of Pharmacology, was also responsible for isolating mescaline as the active psychedelic component from the peyote cactus, thereby initiating a series of studies along with an Institute that, much like the Handbook and the discipline of pharmacology, continues to discover and disseminate new findings to this day. These early endeavors to establish pharmacology as a viable and valuable contributor to the medical sciences met with considerable resistance and challenges. However, the persistence and dedication of these early pharmacologists placed pharmacology on a firm foundation from which to spread this discipline globally, leading ultimately to our current understanding of the principles of drug action and with an impact likely unanticipated by these founding scientists. Summarizing the beginnings of these efforts and their early spread to other countries provides an appropriate context in which to document the many contributions pharmacological research has made over the past 100 years and provide an opportunity to anticipate expectations around its future developments.

Major advances in scientific discovery and insights can result from the development and use of new techniques, as exemplified by the work of Solomon Snyder, who writes a prefatory article in this volume. The Editors have chosen "New Methods and Novel Therapeutic Approaches in Pharmacology and Toxicology" as the Theme for a number of articles in this volume. These include ones that review the development and use of new experimental tools and approaches (e.g., nanobodies and techniques to explore protein-protein interactions), new types of therapeutics (e.g., aptamers and antisense oligonucleotides), and systems pharmacology, which assembles (big) data derived from omics studies together with information regarding drugs and patients. The application of these new methods and therapeutic approaches has the potential to have a major impact on basic and clinical research in pharmacology and toxicology as well as on patient care.

Polyacetylenes are a kind of small active compounds with carbon-carbon triple bond with vast occurrence in plants. Polyacetylenes have attracted considerable attention owing to their diverse biofunctions like tumor suppression, immunity regulation, depression resistance and neural protection. The present review intends to reconstruct data concerning the occurrence, pharmacology, toxicology and pharmacokinetics of polyacetylenes from herbal medicine in a systematic and integrated way, with a view to backing up their curative potential and healthcare properties (2014-2021). The natural polyacetylene-related data were all acquired from the scientific search engines and databases that are globally recognized, such as PubMed, Web of Science, Elsevier, Google Scholar, ResearchGate, SciFinder and CNKI. A total of 183 polyacetylenes were summarized in this paper. Modern pharmacological studies indicated that polyacetylenes possess multiple biological activities including antitumor, immunomodulatory, neuroprotective, anti-depression, anti-obesity, hypoglycemic, antiviral, antibacterial, antifungal, hepatoprotective and renoprotective activities. As important bioactive components of herbal medicine, the pharmacological curative potential of polyacetylenes has been described against carcinomas, inflammatory responses, central nervous system, endocrine disorders and microbial infection in this review. While, further in-depth studies on the aspects of polyacetylenes for toxicity, pharmacokinetics, and molecular mechanisms are still limited, thereby intensive research and assessments should be performed.
