Research has shown that organophosphorus pesticides impair glucose homeostasis and cause insulin resistance and type 2 diabetes. The current study investigates the influence of phoxim on insulin signaling pathways and the protective effects of vitamin E. Phoxim (180 mg kg) and VE (200 mg kg) were administered orally to Sprague-Dawley rats over a period of 28 consecutive days. After exposure to phoxim, the animals showed glucose intolerance and hyperinsulinemia during glucose tolerance tests, and insulin tolerance tests demonstrated an impaired glucose-lowering effect of insulin. Phoxim increases the fasting glucose, insulin and cholesterol levels, as well as the liver hexokinase activity (HK) significantly while decreasing the high density lipoprotein (HDL) cholesterol, and glycogen content in the liver and skeletal muscles observably. Furthermore, we observed an increase of insulin resistance biomarkers and a decrease of insulin sensitivity indices. The insulin receptor substrate (IRS)-2 mRNA expressions of liver and skeletal muscles were down-regulated by phoxim, while the expression of IRS-1 showed no difference. There were no differences in triglycerides, LDL-cholesterol, and fasting glucose treated with phoxim. On the basis of biochemical and molecular findings, phoxim has been determined to impair glucose homeostasis through insulin resistance and insulin signaling pathway disruptions resulting in a reduced function of insulin in hepatocytes and muscles. VE supplementation reduced the fasting glucose, increased the glycogen content and HDL-cholesterol, but did not reduce the insulin resistance indices, when phoxim-treated rats were compared to VE supplemented rats. Overall, this study shows that vitamin E modifies the phoxim toxicity in rats only to a moderate degree.

UDP-glucuronosyltransferases (UGTs) could transform various exogenous and endogenous compounds, which help detoxification of pesticides in insects. To investigate the role of UGTs in the detoxification metabolism of insecticides in Chironomus kiiensis, CkUGT302M1, CkUGT302N1, CkUGT308N1 and CkUGT36J1 genes were identified with 1449-1599 bp encoding 482-532 amino acids. Four UGT genes shared 40.86∼53.36% identity with other homologous insect species, and expressed in all developmental stages, notably in the larval and adult stages. Expression of CkUGTs was higher in the gastric caecum, midgut and head. Moreover, CkUGTs expression and activity were significantly increased in C. kiiensis larvae in exposure to sublethal concentrations of carbaryl, deltamethrin and phoxim, respectively. To further explore the functions of UGT genes, the CkUGT308N1 was effectively silenced in 4th instar C. kiiensis larvae by RNA interference, which resulted in the mortality of dsCkUGT308N1 treated larvae increased by 71.43%, 111.11% and 62.50% under sublethal doses of carbaryl, deltamethrin and phoxim at the 24-h time point, respectively. The study revealed that the CkUGT308N1 gene in C. kiiensis could contribute to the metabolism of pesticides and provide a scientific basis for evaluating the water pollution of pesticides.

Aminoacyl-tRNA synthetases are the key enzymes for protein synthesis. Glycine, alanine, serine and tyrosine are the major amino acids composing fibroin of silkworm. Among them, the genes of alanyl-tRNA synthetase (AlaRS) and glycyl-tRNA synthetase (GlyRS) have been cloned. In this study, the seryl-tRNA synthetase (SerRS) and tyrosyl-tRNA synthetase (TyrRS) genes from silkworm were cloned. Their full length are 1709 bp and 1868 bp and contain open reading frame (ORF) of 1485 bp and 1575 bp, respectively. RT-PCR examination showed that the transcription levels of SerRS, TyrRS, AlaRS and GlyRS are significantly higher in silk gland than in other tissues. In addition, their transcription levels are much higher in middle and posterior silk gland than in anterior silk gland. Moreover, treatment of silkworms with phoxim, an inhibitor of silk protein synthesis, but not TiO NP, an enhancer of silk protein synthesis, significantly reduced the transcription levels of aaRS and content of free amino acids in posterior silk gland, therefore affecting silk protein synthesis, which may be the mechanism of phoxim-silking disorders. Furthermore, low concentration of TiO NPs showed no effect on the transcription of aaRS and content of free amino acids, suggesting that TiO NPs promotes silk protein synthesis possibly by increasing the activity of fibroin synthase in silkworm.

The locusts (Bey-Bienko) and (Zubovski) (Orthoptera Acrididae) and the leaf beetle (Joannis) (Coleoptera, Chrysomelidae) are economically devastating insect species in the desert steppes of Northern China. Control is mainly and frequently dependent on highly toxic chemicals. To date, there have been no complete and comprehensive reports of insecticide applications to these key pests. In this study, laboratory bioassays were carried out to determine and compare the toxicity of twelve insecticides to three outbreaking insects, , , and from three typical desert steppe regions, SZWQ, XHQ and WLTQQ, respectively. The responses of the two locust species and the leaf beetle were evaluated by topical application and leaf dip bioassay techniques across a range of concentrations to develop dosage-mortality regressions. The insecticides tested included six chemical insecticides (β-cypermethrin, imidacloprid, phoxim, λ-cyhalothrin, methomyl, chlorantraniliprole) and six biogenic insecticides (spinosad, avermectin, rotenone, matrine, azadiracthin, and methoxyfenozide). The results showed that phoxim, λ-cyhalothrin, β-cypermethrin and spinosad showed highly toxic activity to , , and , while methonyl, chlorantraniliprole, and rotenone were moderately toxic to both locust species and the leaf beetle. The LC values of matrine, azadiractin, and avermectin were more than 1 μg a.i./adult for and , the LC values of which were higher 2 g/L for . Our findings complement information from previous similar studies and will inform future studies relating to the control of outbreaking insects, such as , , and in desert steppes of northern China. This study is also expected to provide basic data on the use of chemical and biogenic insecticides for application in desert steppes.

(Diptera: Sciaridae) is the most serious root maggot pest which causes substantial damage to the Chinese chive. Organophosphate (OP) and neonicotinoid insecticides are widely used chemical pesticides and play important roles in controlling . However, a strong selection pressure following repeated pesticide applications has led to the development of resistant populations of this insect. To understand the insecticide resistance mechanism in , gene expression analysis might be required. Appropriate reference gene selection is a critical prerequisite for gene expression studies, as the expression stability of reference genes can be affected by experimental conditions, resulting in biased or erroneous results. The present study shows the expression profile of nine commonly used reference genes [, , , , , , , , and )] was systematically analyzed under insecticide stress. Moreover, we also evaluated their expression stability in other experimental conditions, including developmental stages, sexes, and tissues. Five programs (NormFinder, geNorm, BestKeeper, RefFinder, and Δ) were used to validate the suitability of candidate reference genes. The results revealed that the most appropriate sets of reference genes were and across phoxim; and across chlorpyrifos and chlorfluazuron; α and across imidacloprid; α and α across developmental stages; and across larvae; α and across tissues, and and across sex. These results will facilitate the standardization of RT-qPCR and contribute to further research on gene function under insecticides stress.

The 70 kDa heat shock proteins play important roles in protecting organisms against environmental stresses, which are divided into stress-inducible forms (HSP70s) and heat shock cognates (HSC70s). In this study, heat shock protein 70 family was identified in the whole genome of the silkworm. Based on the known nomenclature and phylogenetic analysis, four HSP70s and five HSC70s were classified. Relatively, heat shock cognates were more conservative and were constitutively expressed in various tissues of the silkworm larvae. Under thermal (37 °C and 42 °C) and cold (2 °C) stresses, the expressions of , , and were up-regulated, and the highest induction reached 4147.3, 607.1, and 1987.3 times, respectively. Interestingly, , , and also showed slight induced expressions in the fat body and/or midgut under thermal stresses. In addition, the expression of was induced by dichlorvos and phoxim insecticides, while most HSC70 genes were inhibited. The results suggested that stress-inducible forms play more important roles in adaptation to various stresses than HSC70s.

The poultry red mite Dermanyssus gallinae is the most important blood-sucking ectoparasite in egg-laying hen facilities. The aim of this study was to evaluate the in vitro acaricidal activity of different concentration of authorized (phoxim, ByeMite®, 500 mg/mL) and unauthorized (fipronil, Frontline® 250 mg/100 mL spray) molecules on 14 field isolates of D. gallinae collected from different ornamental poultry farms from different Italian regions. The sensitivity test was performed by contact exposure to four concentrations of each insecticide diluted at 1:5 (10,000-2000-400-80 ppm for phoxim, 500-100-20-4 ppm for fipronil) on a filter paper. The effectiveness of the treatment was significantly (p < 0.0001) associated with the dose of the pesticide used. Considering the mean lethality, phoxim has greater efficacy compared to fipronil (p < 0.001). A great variability of lethality rate was observed with the increase in fipronil dilution; conversely, for phoxim, some outliers were observed, particularly in one farm, suggesting the hypothesis that a certain degree of resistance in the mite population could occur possibly as a consequence of the continual contact with the molecule. This underlines the importance of the use of licensed products administered at correct dosages and the need for alternative molecules to avoid the onset of drug resistance phenomena.

The cotton-melon aphid, Aphis gossypii Glover, is a worldwide-spreading species, and pesticide-resistant populations are increasing rapidly. In this study, investigations were performed based on Illumina HiSeq sequencing of the 16S rDNA V4 region for the bacterial communities embodied as intracellular symbionts under natural and in pesticide-treated populations of A. gossypii. The results revealed that more than 82% of bacterial communities belonged to the phylum Proteobacteria in which the maximum proportion (53.24%) was of the genus Arsenophonus; Hamiltonella composed 22.31; and 1.37% was of the genus Acinetobacter. The relative abundance of Hamiltonella was obvious, vertically transmitted, divided into two groups, and its infection influenced the bacterial communities in A. gossypii. Symbiont density and composition were changed in samples tested on different days. Azadirachtin and phoxim influenced on the composition of bacterial communities. Different biomarkers were used for pesticide-treated samples with LEfSe results. These findings will increase awareness regarding bacterial communities in naturally occurring populations of A. gossypii and pave the way to study the relationship between symbionts and pesticide resistance.

Pesticide residues in aquatic products are of great concern due to the risk of environmental transmission and their extensive use in aquaculture. In our work, a quick screening approach was developed for the qualitative and semi-quantitative screening of 87 pesticide residues in aquatic products. The sample preparation was investigated, including extract solvent, extract methods, buffer salts, lipid removal, cleanup materials and filter membranes for aquatic products. Samples were extracted using a modified QuEChERS procedure, and two clean-up procedures were developed for UHPLC-Q/Orbitrap MS analysis based on the fat content of the aquatic products. The screening detection limits for all studied pesticides were distributed between 1 and 500 μg/kg in the three representative matrices. Seventy-one pesticides could be analyzed with a screening limit between 1 and 25 μg/kg in grass carp and crayfish, sixty-one pesticides could be screened for limits between 1 and 50 μg/kg in crab. The accuracy results showed that recoveries ranged from 50 to 120% for 60, 56 and 52 pesticides at medium-level for grass carp, crayfish and crab, respectively. At high spiking levels, 74, 65 and 59 pesticides were recovered within the range of 50-120% for the three matrices, respectively. The relative standard deviations of most compounds in different matrices were less than 20%. With this method, the local farmed aquatic products were tested for pesticide residues. In these samples, ethoxyquinoline, prometryn and phoxim were frequently detected. The majority of these confirmed compounds did not exceed 2.00 μg/kg. A grass carp with trichlorfon at 4.87 μg/kg and two carps with ethoxyquinoline at 200 µg/kg were detected, indicating the potential dietary risk.

The coix seed is a medicinal and edible plant with rich nutritional and medicinal values. With the expansion of the coix seed consumption market, the problem of coix seed safety has attracted attention worldwide. The aims of this work were to evaluate the contamination of mercury (Hg), lead (Pb), cadmium (Cd), arsenic (As), chromium (Cr) and 116 pesticides in coix seeds collected from 12 main producing regions of coix seeds in the Guizhou Province of China and to analyze the major contributors of heavy metal and pesticide contamination in coix seed. The results show that the average contents of Pb, Cd, As and Cr in the 123 coix seed samples were 0.0069, 0.0021, 0.0138 and 0.1107 mg/kg, respectively, while Hg was not detected in all coix seed samples. Among the five heavy metals detected, only the Cr contents of three samples were found to be higher than the contaminant limit of Chinese standard GB2762-2017 (CSGB). A total of 13 pesticides were detected in 29 samples from seven main production regions of coix seeds, accounting for 23.6% of all the samples. The detection rates of chlorpyrifos were the highest (8.13%), followed by fenpropathrin (4.06%), bifenthrin (2.43%) and phoxim (1.62%), while the detection rates of the remaining pesticides were below 1%. Moreover, the residual risk score of dichlorvos was the highest of all the pesticides detected. The pollution index and risk assessment of heavy metals and pesticide residues indicates that coix seeds were at safe levels for consumption. In the production process of coix seeds, the local government should control the soil in areas heavily polluted by heavy metals and strengthen the monitoring and guidance on the scientific and rational use of pesticides.
