Evaluation of Sensitivity to Phoxim and Cypermethrin in an Endoparasitoid, Meteorus pulchricornis (Wesmael) (Hymenoptera: Braconidae), and Its Parasitization Efficiency Under Insecticide Stress.
Insecticides can have consequences for beneficial arthropods. Insect parasitoids can contact insecticides through direct exposure spray droplets or residues on crop foliage. Here, we focus on better understand the response of Meteorus pulchricornis (Wesmael), a parasitoid wasp of lepidopteran pests, and its detoxification mechanisms on stress caused by phoxim and cypermethrin. Hence, we determined the dose-mortality curves and estimating the sublethal concentrations (LC30 and LC50). Then, we applied the sublethal concentrations against adult parasitoids to assess its survival, parasitism efficacy, and also developmental and morphometric parameters of their offspring. Simultaneously, we check the activities of glutathione S-transferase (GST), acetylcholinesterase (AChE), and peroxidase (POD) after sublethal exposure of both insecticides, which has measured until 48 h after treatment. Overall, phoxim and cypermethrin exhibited acute lethal activity toward the parasitoid with LC50 values 4.608 and 8.570 mg/liter, respectively. Also, we detect that LC30 was able to trigger the enzymatic activity of GST, AChE, and POD, suggesting a potential detoxification mechanism. However, even when subjected to sublethal exposure, our results indicate strong negatives effects, in particular for phoxim, which has affected the parasitism efficacy and also the developmental and morphometric parameters of M. pulchricornis offspring. Therefore, it can be concluded that both phoxim and cypermethrin have negative impacts on M. pulchricornis and we suggest cautioning their use and the need for semifield and field assessments to confirm such an impact.

BACKGROUND
Bedbugs are nocturnal ectoparasites that have made a comeback after 20 years and have become one of the main challenges for pest control methods worldwide. Monitoring chemical pesticide resistance is crucial for identifying the best bed bugs management strategies to effectively manage arthropods. This study aims to assess the susceptibility of (Hemiptera: Cimicidae) collected from different parts of Khorasan-Razavi Province, (northeast of Iran) to deltamethrin (pyrethroid), phoxim (organothiophosphate) and propetamphos (phosphoramidate).
METHODS
This study was conducted from Dec 2020 to May 2021. The efficacy of three insecticides (deltamethrin, phoxim, and propetamphos) on adult was assessed using a bioassay method recommended by the World Health Organization (WHO),. Concentrations of deltamethrin used were 10, 20, 40, 80, 160, 320, 640, 1280, and 2560 ppm, while concentrations of phoxim and propetamphos were 10, 40, 160, and 320 ppm. The bed bugs were continuously exposed to the insecticide for 24 hours, and mortality was assessed at regular intervals during the observation period. The concentration-response data were subjected to POLO-PC software and data were analyzed by the one-way and two-way ANOVA procedures.
RESULTS
The lethal concentration fifty values of deltamethrin, phoxim and propetamphos on the examined bed bugs were 0.551, 0.148 and 0.237 ppm, respectively. Insecticide effects of phoxim were significantly higher (P≤ 0.05) compared to each of either deltamethrin and propetamphos agents.
CONCLUSION
The insecticide effects of phoxim against bed bugs were significantly higher compared to each of either deltamethrin or propetamphosinsecticides.

Due to overuse and terrestrial input, there are large quantities of phoxim and prometryne residues in some aquatic environments. In the present study, the effects of these compounds on Penaeus vannamei hepatopancreas were analysed at the transcriptome level to investigate toxicity in this nontarget aquaculture organism. Twelve normalised cDNA libraries were constructed using RNA from phoxim and prometryne treatment groups, and an untreated control group. A total of 667,750,902 clean reads were obtained. Analysis of differentially expressed genes (DEGs) identified 449 in control vs phoxim groups, 185 in control vs prometryne groups, and 183 in prometryne vs phoxim groups. In the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, arachidonic acid metabolism, pancreatic secretion, linoleic acid metabolism, and beta-alanine metabolism pathways were significantly enriched in control vs phoxim groups. In control vs prometryne groups, lysosome, pentose and glucuronate interconversion, antigen processing and presentation, and glycosaminoglycan degradation pathways were significantly enriched. In prometryne vs phoxim groups, protein digestion and absorption, extracellular matrix (ECM)-receptor interaction, PI3K-Akt signalling, cell adhesion molecule (CAM), AGE-RAGE signalling related to diabetic complications, focal adhesion, and renin secretion pathways were significantly enriched. In further detailed analysis, glutathione S-transferase (GST), glutathione peroxidase and basic phospholipase A2 were downregulated in the phoxim treatment group, indicating that phoxim damaged hepatopancreas. Upregulation of phospholipase A2 (secretory phospholipase A2-like) indicates possible inflammatory pathological injury to hepatopancreas caused by phoxim. Meanwhile, downregulation of CD63 indicates that prometryne affect the immune system.

The poultry red mite Dermanyssus gallinae is the most important blood-sucking ectoparasite in egg-laying hen facilities. The aim of this study was to evaluate the in vitro acaricidal activity of different concentration of authorized (phoxim, ByeMite®, 500 mg/mL) and unauthorized (fipronil, Frontline® 250 mg/100 mL spray) molecules on 14 field isolates of D. gallinae collected from different ornamental poultry farms from different Italian regions. The sensitivity test was performed by contact exposure to four concentrations of each insecticide diluted at 1:5 (10,000-2000-400-80 ppm for phoxim, 500-100-20-4 ppm for fipronil) on a filter paper. The effectiveness of the treatment was significantly (p < 0.0001) associated with the dose of the pesticide used. Considering the mean lethality, phoxim has greater efficacy compared to fipronil (p < 0.001). A great variability of lethality rate was observed with the increase in fipronil dilution; conversely, for phoxim, some outliers were observed, particularly in one farm, suggesting the hypothesis that a certain degree of resistance in the mite population could occur possibly as a consequence of the continual contact with the molecule. This underlines the importance of the use of licensed products administered at correct dosages and the need for alternative molecules to avoid the onset of drug resistance phenomena.

Photocatalytic degradation of commercial phoxim emulsion in aqueous suspension was investigated by using La-doped mesoporous TiO2 nanoparticles (m-TiO2) as the photocatalyst under UV irradiation. Effects of La-doping level, calcination temperature, and additional amount of the photocatalyst on the photocatalytic degradation efficiency were investigated in detail. Experimental results indicate that 20 mg L(-1) phoxim in 0.5 g L(-1) La/m-TiO2 suspension (the initial pH 4.43) can be decomposed as prolonging the irradiation time. Almost 100% phoxim was decomposed after 4 h irradiation according to the spectrophotometric analyses, whereas the mineralization rate of phoxim just reached ca. 80% as checked by ion chromatography (IC) analyses. The elimination of the organic solvent in the phoxim emulsion as well as the formation and decomposition of some degradation intermediates were observed by high-performance liquid chromatography-mass spectroscopy (HPLC-MS). On the basis of the analysis results on the photocatalytic degradation intermediates, two possible photocatalytic degradation pathways are proposed under the present experimental conditions, which reveal that both the hydrolysis and adsorption of phoxim under UV light irradiation play important roles during the photocatalytic degradation of phoxim.

In this work, we reported the first use of a novel fluorescent probe for phoxim detection based on yellowish-green emissive carbon quantum dots (CQDs). By facile one-pot hydrothermal treatment of citric acid and p-phenylenediamine, the CQDs with yellowish-green luminescence were synthesized. The sensing mechanism is based on the quenching effect of phoxim on the fluorescence emission of CQDs by inner filter effect (IFE). The CQDs showed a good sensitivity and selectivity to phoxim detection and dual good linear relationships were provided in the concentration ranges of 1-10 μM and 10-98 μM, associated with the limit of detection of 0.09 μM. The proposed assay has been allowed to detect phoxim in real samples with satisfying recoveries ranging from 93 to 105.9%. Furthermore, due to the low cytotoxicity and excellent biocompatibility, the as-prepared CQDs was successfully used for cell imaging, demonstrating the considerable potential applications of such CQDs in biological field.

Phoxim is a useful organophosphate (OP) pesticide used in agriculture in China, however, exposure to this pesticide can result in a significant reduction in cocooning in Bombyx mori (B. mori). Titanium dioxide nanoparticles (TiO2 NPs) have been shown to decrease phoxim-induced toxicity in B. mori; however, very little is known about the molecular mechanisms of silk gland damage due to OP exposure and repair of gland damage by TiO2 NP pretreatment. In the present study, exposure to phoxim resulted in a significant reduction in cocooning rate in addition to silk gland damage, whereas TiO2 NP attenuated phoxim-induced gland damage, increased the antioxidant capacity of the gland, and increased cocooning rate in B. mori. Furthermore, digital gene expression data suggested that phoxim exposure led to significant alterations in the expression of 833 genes. In particular, phoxim exposure caused significant down-regulation of Fib-L, Ser2, Ser3, and P25 genes involved in silk protein synthesis, and up-regulation of SFGH, UCH3, and Salhh genes involved in silk protein hydrolysis. A combination of both phoxim and TiO2 NP treatment resulted in marked changes in the expression of 754 genes, while treatment with TiO2 NPs led to significant alterations in the expression of 308 genes. Importantly, pretreatment with TiO2 NPs increased Fib-L, Ser2, Ser3, and P25 expression, and decreased SFGH, UCH3, and Salhh expression in silk protein in the silk gland under phoxim stress. Therefore, Fib-L, Ser2, Ser3, P25, SFGH, UCH3, and Salhh may be potential biomarkers of silk gland toxicity in B. mori caused by phoxim exposure.

A ratiometric electrochemical immunosensor, based on DNA tetrahedron nanostructure (DTNS), is introduced for vegetable phoxim determination. DTNS spontaneously adheres onto gold-nanoparticle-modified electrode and forms stable three-dimensional structure, providing plenty of binding sites to the built-in reference, methylene blue (MB). Monoclonal antibody (m-Ab) is vertically linked onto DTNS vertex, selectively responses antigenic phoxim, and promotes the target signal of I. Thus, a ratiometric indicator, I/I, is sensibly established with the target signal (I) and the reference signal (I). Modifications, mechanisms and advances of the proposed method are subsequently examined with morphological methods and electrochemical experiments. This method brings considerable advances in analytical behaviors. The ratiometric signal presents better performance than solo system in repeatability and long-time stability. As-fabricated sensor presents wide dynamic range as 0.1∼30 μg/L, and limit of detection is well defined as 0.003 μg/L (S/N = 3). Finally, this method is verified with real-vegetable-sample analysis, certified HPLC and recovery test.

BACKGROUND
Fleas (Ceratophyllus sciurorum) are common on farmed mink in Denmark. When present, the fleas have a negative impact on the health of the farmed mink and are of nuisance for farm staff. Severe infestations of fleas cause anemia, poor growth and may result in death of mink kits. Changed behavior of the dams is also observed. Further it has been demonstrated that the fleas are vectors of Aleutian disease virus. Flea control is based on use of a few insecticides and resistance has been reported against permethrin. There is thus a need for new flea control products. In this blinded, randomized clinical trial according to GCP standard, phoxim spray and bendiocarb powder for flea control on mink farms were investigated.
RESULTS
Both the phoxim spray solution and bendiocarb powder were found to be efficient for the control of C. sciurorum fleas on farmed mink. Phoxim treatments reduced the number of fleas by 98.4% and the bendiocarb treatments reduced the number of fleas by 99.0% in the mink nest boxes when compared to counts in controls. No clinical signs were observed post treatment.
CONCLUSIONS
The study demonstrated that phoxim sprayed on the animals and the use of bendiocarb powder in the nest box material were highly efficient for the control of the C. sciurorum fleas on farmed mink. Both products were safe to use at the recommended dose rate. Both compounds are recommended to be integrated in a new farm management plan suggested here.

Phoxim (emulsifiable concentrate (EC) and granules (G)) has been widely used in bamboo forests. The persistence and magnitude of phoxim residues in the crop and soil must be investigated to ensure human and environmental safety. The environmental behaviors of the two formulations were investigated in a bamboo forest under soil surface mulching conditions (CP) and non-covered cultivation conditions (NCP). The half-lives of phoxim in soil under the two conditions in soil were 4.1-6.2days (EC) and 31.5-49.5days (G), respectively. Phoxim in EC could be leached from the topsoil into the subsoil. A minimized leaching effect was observed for G under NCP. Inversely, an enhanced leaching effect was observed for G under CP. The G formulation resulted in more parent compound (in bamboo shoots) and metabolite (in soil) residues of phoxim than in the case of EC, especially under CP conditions. In addition, the intensity and duration of the formulation effect on soil pH adjustment from G were more obvious than that from EC. Results showed that the environmental behaviors (distribution, degradation, residue) of phoxim in the bamboo forest were significantly influenced by the type of formulation. The prolongation effect from phoxim G might cause persistence and long-term environmental risk. However, bamboo shoot consumption could be considered relatively safe after applying the recommended dose of the two phoxim formulations.
