Recently, various efforts have been made to explore the potential of natural polysaccharides derived from sea weeds to promote sustainable development. Herein, carrageenan (CG), a polysaccharide extracted from red sea algae, was utilized to design network structures as hydrogels, aimed at significant applications in drug delivery (DD) systems. Hydrogels were designed by graft copolymerization reaction of poly(bis [2-methacryloyloxy] ethyl phosphate [poly(BMEP)] and poly(acrylic acid) [poly(AAc)] onto CG in the presence of a crosslinking agent. Hydrogels were developed by covalent linkage by graft copolymerization and supramolecular interactions, existing in the copolymers. Copolymers were characterized by Atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR), C-nuclear magnetic resonance (NMR), and X-ray diffraction (XRD) instrumentations. The drug diffusion exhibited a sustained pattern due to polymer-drug interactions. The drug release followed non-Fickian diffusion mechanism and the release profile was most accurately depicted by first order kinetic model. The biocompatible nature of the copolymer was demonstrated from the hemolytic index value signifying minimal adverse interactions with blood component upon exposure. A protein adsorption test was performed using bovine serum albumin (BSA), exhibiting 8.15 ± 0.26 % albumin adsorption. Polymers exhibited mucoadhesive character, evidenced by their requirement of a detachment force measuring 195 ± 4.72 mN for separation from the membrane during interactions with the mucosal surface. The hydrogels exhibited antioxidant properties, evidenced by 2, 2'-Diphenylpicrylhydrazyl (DPPH) assay, revealing copolymer capable of scavenging 58.21 ± 2.26 % of free radical. The hydrogel revealed antimicrobial activity against P. aeruginosa and S. aureus bacteria, a property further enhanced in hydrogels with the drug doxycycline. These findings suggest suitability of these hydrogels for biomedical applications, with a significant emphasis on drug delivery.

There are large areas of saline-alkaline waters worldwide, the utilization of which would greatly enhance the development of aquaculture productivity. To elucidate the regulatory mechanisms underlying the adaptation of large yellow croaker (Larimichthys crocea) to saline-alkaline water, this study analyzed the growth performance, tissue histology, and gills transcriptome profiles of L. crocea in both seawater (CK) and saline-alkaline water (EX) groups. Growth indices statistics revealed that L. crocea can adapt to saline-alkaline water, with growth performance comparable to that of the CK group. Histological examination revealed partial cellular detachment and structural relaxation in the gills tissue of the EX group, while liver and kidney tissues appeared normal. Transcriptome analysis revealed 3821 differentially expressed genes (DEGs), with 1541 DEGs up-regulated and 2280 DEGs down-regulated. GO enrichment analysis indicated that up-regulated DEGs were enriched in terms related to metabolite production during biological activities, while down-regulated DEGs were associated with terms related to maintaining cellular activities. KEGG enrichment analysis revealed that up-regulated DEGs were enriched in pathways related to the synthesis and metabolism of amino acids and lipids, such as the PPAR signaling pathway and glutathione metabolism. The down-regulated DEGs were predominantly enriched in immune-related signaling pathways, including the Toll-like receptor signaling pathway and NOD-like receptor signaling pathway. Further analysis revealed that genes such as lipoprotein lipase A (lpla), branched-chain amino acid aminotransferase 2 (bcat2), interleukin 8 (il8), interleukin 10 (il10), and interferon regulatory factor 7 (irf7) were involved in the adaptation of L. crocea to saline-alkaline water culture conditions. This study provides a basis for understanding the adaptability of large yellow croaker to saline-alkaline water and lays the foundation for the rational utilization of fishery water resources.

Liposomes (LIP) loaded with natural active ingredients have significant potential in the food industry. However, their low loading efficiency (LE) hampers the advancement of liposomal products. To improve the loading capacity of functional compounds, bionic oleosomes (BOLE) with a monolayer of phospholipid membranes and a glyceryl tricaprylate/caprate (GTCC) oil core have first been engineered by high-pressure homogenization. TEM revealed that the core of BOLE consists of GTCC instead of water, thereby extending the hydrophobic space. Steady-state fluorescence and active loading experiments confirmed that cholesterol (CH) detached from the phospholipid membrane and entered the oil core, where it repelled cannabidiol (CBD). Based on the extending hydrophobic space, CBD-BOLE was prepared and its LE was 3.13 times higher than CBD-LIP. The CBD-phospholipid ratio (CPR) of CBD-BOLE significantly improved at least 7.8 times. Meanwhile, the free radical scavenging activity of CBD was increased and cytotoxicity was reduced.

Most multiplexed photoelectrochemical (PEC) sensors require additional instrumentation and cumbersome electrode modification and surface partitioning, which limits their portability and instrument miniaturization. Herein, a pH-responsive programmable triple DNA nanomachine was developed for constructing a reconfigurable multiplex PEC sensing platform. By programming the base sequence, T-A·T-riched triple DNA was designed to construct integrated nano-controlled release machine (INCRM) for simultaneous recognition of multiple targets. The INCRM enables to recognize two targets in one step, and sequentially separate the signal labels by pH adjustment. The detached signal label catalyzes glucose to produce gluconic acid, causing the C-riched DNA fold into a triple structure on the electrode surface. As a result, one target can be detected relying on the enhanced photocurrent due to accelerated electron transfer between the CdS QD labeled at the end of C-riched DNA and the electrode. The triplex DNA dissociation in pH 7.4 buffer reconfigures the electrode interface, which can be continued to detect another target. The feasibility of the multiplexed sensor is verified by the detection of extensively coexisting antibiotics enrofloxacin (ENR) and ciprofloxacin (CIP). Under the optimal conditions, wide linear range (10 fg/mL ∼ 1 μg/mL) and low detection limit (3.27 fg/mL and 9.60 fg/mL) were obtained. The pH-regulated programmable triplex DNA nanomachine-based sensing platform overcomes the technical difficulties of conventional multiplexed PEC assay, which may open the way for miniaturization of multiplexed PEC sensors.

The proliferation and death of granulosa cells (GCs) in poultry play a decisive role in follicular fate and egg production. The follicular fluid (FF) contains a variety of nutrients and genetic substances to ensure the communication between follicular cells. Exosomes, as a new intercellular communication, could carry and transport the proteins, RNA, and lipids to react on GCs, which had been found in FF of various domestic animals. Whether exosomes of FF in poultry play a similar role is unclear. In this study, geese, a poultry with low egg production, were chosen, and the effect of FF exosomes on the proliferation and death of GCs was investigated. Firstly, there were not only a large number of healthy small yellow follicles (HSYFs) but also some atresia small yellow follicles (ASYFs) in the egg-laying stage. Also, the GC layers of ASYFs became loose interconnections, inward detachment, and diminished survival rate than that of HSYFs. Besides, compared to HSYFs, the contents of E2, P4, and the mRNA expression levels of ferroptosis-related genes GPX4, FPN1, and FTH1 were significantly decreased, while COX2, NCOA4, VDAC3 mRNA were significantly increased, and the structure of mitochondrial cristae disappeared and the outer membrane broke in the GC layers of ASYFs. Moreover, the ROS, MDA, and oxidation levels in the GC layers of ASYFs were significantly higher than those of HSYFs. All these hinted that ferroptosis might result in a large number of GCs death and involvement in follicle atresia. Secondly, FF exosomes were isolated from HSYFs and ASYFs, respectively, and identified by TEM, NTA, and detection of exosome marker proteins. Also, we found the exosomes were phagocytic by GCs by tracking CM-Dil. Moreover, the addition of ASYF-FF exosomes significantly elevated the MDA content, Fe levels, and the mitochondrial membrane potential (MMP) in GCs, thus significantly inhibiting the proliferation of GCs, which was restored by the ferroptosis inhibitor ferrostatin-1. Thirdly, the proteomic sequencing was performed between FF-derived exosomes of HSYFs and ASYFs. We obtained 1615 differentially expressed proteins, which were mainly enriched in the protein transport and ferroptosis pathways. Among them, HMOX1 was enriched in the ferroptosis pathway based on differential protein-protein interaction network analysis. Finally, the role of HMOX1 in regulating ferroptosis in GCs was further explored. The highly expressed HMOX1 was observed in the exosomes of ASYF-FF than that in HSYF-FF. Overexpression of HMOX1 increased ATG5, LC3II, and NCOA4 expression and reduced the expression of FTH1, GPX4, PCBP2, FPN1 in the ferroptosis pathway, also promoted intracellular Fe accumulation and MDA surge, which drove ferroptosis in GCs. The effects of HMOX1 on ferroptosis could be blocked by its inhibitor Znpp. Taken together, the important protein HMOX1 was identified in FF, which could be delivered to GCs via exosomes, triggering ferroptosis and thus determining the fate of follicles.

Triclocarban (TCC), an antibacterial agent commonly used in personal care products, is one of the top ten contaminants of emerging concern in various environmental media, including soil and contaminated water in vadose zone. This study aimed to investigate TCC-contaminated water remediation using biochar-immobilized bacterial cells. Pseudomonas fluorescens strain MC46 (MC46), an efficient TCC-degrading isolate, was chosen, whereas agro-industrial carbonized waste as biochar was directly used as a sustainable cell immobilization carrier. According to the long-term TCC removal performance results (160 d), the biochar-immobilized cells consistently exhibited high TCC removal efficiencies (84-97%), whereas the free MC46 removed TCC for 76-94%. At 100 days, the detachment of the MC46 cells from the immobilized cell column was observed. The micro-Fourier-transform infrared spectroscopy results indicated that extracellular polymeric substance (EPS) was produced, but polysaccharide and protein fractions were washed out of the column. The lipid fraction of EPS adhered to the biochar, promoting TCC sorption for long-term treatment. The shortening of MC46 cells improved the tolerance of TCC toxicity. The TCC-contaminated water was successfully detoxified by the biochar-immobilized MC46 cells. Overall, the waste-derived biochar-immobilized cell system proposed in this study for the removal of emerging contaminants, including TCC, is efficient, economical, and aligned with the sustainable development concept of value-added utilization of waste.

Terminal deletions of chromosome 3q are associated with a heterogenous clinical phenotype, which includes growth restriction, developmental delay, and intellectual disability. However, little has been published on the ophthalmic impacts of chromosome 3q deletions. We report a 9-year-old boy with a 1.4 megabase deletion of 3q27.1q27.2 whose ocular morbidities included retinal detachment in one eye, vitreous hemorrhage in the other eye, and foveal hypoplasia in both eyes that required acute care and continuous ophthalmologic follow-up.

Ultrasonic technology has a significant degassing effect and can increase the efficiency of hydrogen production in the proton exchange membrane electrolysis of water. However, further research is needed to understand its influence mechanism on hydrogen bubbles. In this work, a kinetic analysis is performed to investigate the principle of hydrogen production and the kinetic behaviour of hydrogen bubble evolution by applying the ultrasonic amplification technique under static and flow dynamics in the proton exchange membrane electrolysis cell. The evolution of hydrogen bubbles in the static and in the flow dynamic of the aqueous electrolyte solution under ultrasound was characterised by imaging. The results show that the aqueous electrolyte solution in the flow state reduces the size of hydrogen bubbles and increases the detachment speed compared to the static state, which promotes the process of hydrogen bubble evolution, and that the thermal effect of ultrasound on the temperature of the aqueous electrolyte solution in the flow state is very small compared to the static state and can be ignored. Ultrasound has different effects on the different stages of hydrogen bubble evolution. In the nucleation stage, the ultrasonic cavitation effect increases the highly reactive radicals such as •OH, H•, etc., and the mechanical vibration effect of ultrasound increases the nucleation sites, which are denser and more evenly distributed. In the growth phase, the ultrasonic cavitation effect and the mechanical vibration effect promote the breaking of hydrogen bonds of water molecules and improve mass transport, which promotes the growth of hydrogen bubbles, and the fluctuating energy of positive and negative ultrasound promotes the growth of hydrogen bubbles with the vibration speed. In the detachment phase, the radius of the hydrogen bubbles is influenced by the ultrasound. The radius of the hydrogen bubbles changes with the positive and negative ultrasonic pressure, the radius of the hydrogen bubbles at negative ultrasonic pressure increases, the positive ultrasonic pressure decreases, the changing effect of the radius of the hydrogen bubbles favours the detachment of the hydrogen bubbles. In the polymerisation phase, the ultrasound leads to increased polymerisation of the fine bubble streams. Ultrasound contributes to the hydrogen production effect of proton exchange membrane water electrolysis in actual operation.

RATIONALE
Complete dislodgement of a mechanical valve is extremely uncommon as a long-term issue after getting a substitute mitral valve, and this report details an incident of complete detachment of a mechanical valve.
PATIENT CONCERNS
A 50-year-old woman, who underwent mitral mechanical valve replacement 2 decades earlier at another facility, was urgently admitted due to sudden cardiogenic shock.
DIAGNOSES
Transthoracic echocardiograms revealed severe malfunction of the mitral valve prosthesis, characterized by significant mitral regurgitation and moderate pulmonary hypertension. Following the insertion of extracorporeal membrane oxygenation and an intra-aortic balloon pump, the hemodynamics stabilized. Coronary angiography displayed the prosthetic mitral valve ring and leaflet floating in the left atrium, as confirmed by preoperative real-time 3-dimensional transesophageal echocardiography. A complete separation of the prosthetic ring and leaflet from the suture ring was observed.
INTERVENTIONS
The patient promptly underwent bioprosthetic mitral valve replacement.
OUTCOMES
The patient's postoperative course was uneventful, leading to discharge in good condition.
LESSONS
A crucial aspect is comprehending the structure of the prosthetic valve itself. The use of transthoracic echocardiography and real-time 3-dimensional transesophageal echocardiography provides additional structural and functional details, enhancing support for potential life-saving interventions. Echocardiography plays a significant role in evaluating the morphology and function of prosthetic valves.

DNA damage is one of the most important effects induced by chemical agents. We report a comparative analysis of DNA fragmentation on three different cell lines using terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay, generally applied to detect apoptosis. Our approach combines cytogenetic techniques and investigation in detached cellular structures, recovered from the culture medium with the aim to compare the DNA fragmentation of three different cell line even beyond the cells adherent to substrate. Consequently, we detect any fragmentation points on single chromosomes, whole nuclei and other cellular structures. Cells were exposed to resveratrol (RSV) and doxorubicin (Doxo), in single and combined treatments. Control and treated astrocytes showed DNA damage in condensed nuclei and detached structures. Caco-2 cells showed fragmented DNA only after Doxo-treatment, while controls showed fragmented chromosomes, indicating DNA damage in replicating cells. MDA-MB-231 cells showed nuclear condensation and DNA fragmentation above all after RSV-treatment and related to detached structures. This model proved to perform a grading of genomic instability (GI). Astrocytes show a hybrid level of GI. Caco-2 cells showed fragmented metaphase chromosomes, proving that the DNA damage was transmitted to the daughter cells probably due to an absence of DNA repair mechanisms. Instead, MDA-MB-231 cells showed few or no fragmented metaphase, suggesting a probable activation of DNA repair mechanisms. By applying this alternative approach of TUNEL test, we obtained data that can more specifically characterize DNA fragmentation for a suitable application in various fields.
