Eicosanoids, which are oxygenated derivatives of polyunsaturated fatty acids (PUFAs), serve as signaling molecules that regulate spermatogenesis in mammals. However, their roles in crustacean sperm development remain unknown. In this study, the testis and vas deferens of the black tiger shrimp Penaeus monodon were analyzed using ultra-high performance liquid chromatography coupled with Orbitrap high resolution mass spectrometry. This led to the identification of three PUFAs and ten eicosanoids, including 15-deoxy-Δ12,14-prostaglandin J2 (15d-PGJ2) and (±)15-hydroxyeicosapentaenoic acid ((±)15-HEPE), both of which have not previously been reported in crustaceans. The comparison between wild-caught and domesticated shrimp revealed that wild-caught shrimp had higher sperm counts, higher levels of (±)8-HEPE in testes, and higher levels of prostaglandin E2 (PGE2) and prostaglandin F2α in vas deferens than domesticated shrimp. In contrast, domesticated shrimp contained higher levels of (±)12-HEPE, (±)18-HEPE, and eicosapentaenoic acid (EPA) in testes and higher levels of 15d-PGJ2, (±)12-HEPE, EPA, arachidonic acid (ARA), and docosahexaenoic acid (DHA) in vas deferens than wild-caught shrimp. To improve total sperm counts in domesticated shrimp, these broodstocks were fed with polychaetes, which contained higher levels of PUFAs than commercial feed pellets. Polychaete-fed shrimp produced higher total sperm counts and higher levels of PGE2 in vas deferens than pellet-fed shrimp. In contrast, pellet-fed shrimp contained higher levels of (±)12-HEPE, (±)18-HEPE, and EPA in testes and higher levels of (±)12-HEPE in vas deferens than polychaete-fed shrimp. These data suggest a positive correlation between high levels of PGE2 in vas deferens and high total sperm counts as well as a negative correlation between (±)12-HEPE in both shrimp testis and vas deferens and total sperm counts. Our analysis not only confirms the presence of PUFAs and eicosanoids in crustacean male reproductive organs, but also suggests that the eicosanoid biosynthesis pathway may serve as a potential target to improve sperm production in shrimp.

AIM
OvSynch is a hormonal protocol for synchronization of estrus and use of artificial insemination (AI) at an optimal time without adverse effects on the ovaries or uterus. This study investigated the use of noninvasive color Doppler ultrasound to assess changes in uterine and vaginal blood flow during the Ovsynch program for synchronization of estrus and its relation to the pregnancy rates in Holstein cows.
MATERIALS AND METHODS
The experimental cows received an intramuscular dose of 10 μg of a GnRH analogue (G1), followed 7 days later with an intramuscular injection of synthetic prostaglandin F2α (P: PGF2α) analogue (500 μg cloprostenol sodium), and given a 10 μg, injection of the GnRH analogue (G2) i.m. 48 h after the PGF2α treatment, and the cows were bred 14-16 h after. Uterine and vaginal perfusion were investigated by performing transrectal Doppler ultrasonography of both the uterine and vaginal arteries in Holstein cows at different time points during the Ovsynch program to determine: peak systolic velocity (PSV), time-averaged maximum velocity (TAMV), the volume of blood flow (BFV), pulsatility index (PI), resistance index (RI), resistance impedance (S/D) and diameters of uterine (UA) and vaginal (VA) arteries. Steroid hormones were also assayed. Transrectal ultrasonography (TUS) was performed at 32 and 60 days to confirm the pregnancy per artificial insemination (P/AI).
RESULTS
The uterine PSV, TAMV, and PV were greater at the time of the cloprostenol sodium and second GnRH injections (p<0.05) than at the time of the first GnRH injection. The vaginal PSV, PV were greater at the time of the cloprostenol sodium than at the time of the first and second GnRH injections (p<0.05). The receiver operating characteristic curve (ROC curve) indicated a high correlation between the uterine and vaginal blood flow and the rate of the pregnancy (p<0.05). The area under the ROC curve was 0.920 and 0.87 (p<0.05) for vaginal and uterine arteries respectively at time of G2. The serum levels of progesterone, estrogen and cortisol were correlated with the P/AI (p<0.05). The P/AI significantly decreased from 43.9 % at 32 d to 35.37 % at 60 d.
CONCLUSION
These results indicate that noninvasive Doppler ultrasonography is a valid method to evaluate changes in the characteristics of uterine and vaginal blood flow in cows during the Ovsynch protocol. Furthermore, vaginal and uterine blood flow are two determinant factors for the higher conception rates in Holstein dairy cows.

This research aimed to determine the effect of temperament on reproductive parameters including cortisol and progesterone (P4) in Nellore cows. Additionally, two methods for increasing plasma progesterone (P4) levels in excitable animals to enhance pregnancy rate (P/AI) and reduce pregnancy loss were investigated. In total, 939 cows were subjected to timed artificial insemination (TAI) and divided into three groups: (P4LA; n = 305) 150 mg of injectable long-acting progestogen 7 days after TAI; (GnRH; n = 306), 10 μg of buserelin acetate on day 7 after TAI; control group (CG; n = 328) without hormonal treatment. In 213 cows, randomly chosen from each group, ultrasound evaluations of the preovulatory follicle (Mode B) were performed on the day of insemination and of the corpus luteum (Color Doppler) 7 and 16 days after TAI. Blood samples were obtained from 20% of the 939 animals, randomly chosen from each group, on the day of insemination and after 7 and 16 days to measure cortisol and progesterone, respectively. At the time of insemination, subjective temperament evaluations were performed with the animals being classified as excitable (EXC) or adequate (ADQ). The SAS GLIMMIX procedure was used to compare the pregnancy rate (P/AI) and gestational loss within each temperament for the three experimental groups. Continuous variables were analyzed utilizing SAS PROC MIXED procedure. Cortisol concentration was higher and POF (preovulatory follicle) and CL (corpus luteum) volumes at the time of insemination and 7 days after AI, respectively, were lower in EXC animals than in ADQ. No significant difference was observed between the number of pixels, CL intensity, and plasma concentration of P4, 7 days after TAI. However, 16 days post-insemination, among the animals classified as EXC, higher concentrations of P4 were observed in the GnRH and P4LA groups than in the control. Regarding P4 concentrations, there was a tendency to be lower in animals classified as EXC than in ADQ within the control group (P = 0.06), while rate of blood flow from the CL was lower in EXC animals than in ADQ animals (P = 0.04). Among the ADQ animals, the GnRH and P4LA groups showed a lower CL flow rate than that observed in the control (P = 0.04), 16 days after the TAI. Among EXC animals, a higher pregnancy rate was observed in the GnRH and P4LA groups than in the control group (P = 0.01). In the control group, the pregnancy rate (P/AI) of the ADQ animals was higher than that of the EXC animals (P = 0.05). No statistically significant differences were observed between gestational losses when the treatments or temperaments were compared. In conclusion, the use of GnRH or P4LA, 7 days after insemination, improves pregnancy rates in excitable animals and is a viable alternative to minimize the negative impact of stress and improve reproductive efficiency in beef cattle.

Effect of a targeted reproductive management program designed to prioritize insemination at detected estrus and optimize time to insemination on the reproductive performance of lactating dairy cows.
The primary objective of this randomized controlled experiment was to evaluate the insemination dynamic and reproductive performance of cows managed with a targeted reproductive management (TRM) program designed to prioritize artificial insemination (AI) at detected estrus (AIE) and optimize timing of AI by grouping cows based on detection of estrus during the voluntary waiting period (VWP). Our secondary objective was to evaluate reproductive outcomes for cows with or without estrus during the VWP. Lactating Holstein cows fitted with an ear-attached sensor for detection of estrus were randomly assigned to a TRM treatment that prioritized AIE based on detection of estrus during the VWP (TP-AIE; n = 488), a non-TRM treatment that prioritized AIE (P-AIE; n = 489), or an all timed AI (TAI) treatment with extended VWP (ALL-TAI; n = 491). In TP-AIE, cows with or without automated estrus alerts (AEA) recorded during the VWP received AIE if detected in estrus for at least 31 ± 3 or 17 ± 3 d after a 49 d VWP, respectively. Cows not AIE with or without AEA during the VWP received TAI after Ovsynch with progesterone supplementation and 2 PGF treatments (P4-Ov) at 90 ± 3 or 74 ± 3 d in milk (DIM), respectively. In P-AIE, cows received AIE if detected in estrus for 24 ± 3 d after a 49 d VWP, and if not AIE received TAI at 83 ± 3 DIM after P4-Ov. In ALL-TAI, cows received TAI at 83 ± 3 DIM after a Double-Ovsynch protocol. Data were analyzed by logistic and Cox's proportional hazard regression. The proportion of cows AIE did not differ for TP-AIE (71.0%) and P-AIE (74.6%). Overall P/AI at 39 d after first service was greater for the ALL-TAI (47.6%) than for the P-AIE (40.2%) and TP-AIE (39.5%) treatments. The hazard of pregnancy up to 150 DIM was greater for cows in TP-AIE (hazard ratio = 1.2; 95% confidence interval: 1.1-1.4) and P-AIE (hazard ratio = 1.2; 95% confidence interval: 1.1-1.4) than for cows in the ALL-TAI treatment which resulted in median time to pregnancy of 89, 89, and 107 d. Conversely, the proportion of cows pregnant at 150 DIM did not differ (ALL-TAI 78.5%, P-AIE 76.3%, TP-AIE 76.0%). Except for a few outcomes for which no difference was observed, cows detected in estrus during the VWP had better performance than cows not detected in estrus. Cows with AEA during the VWP were more likely to receive AIE, had greater P/AI, and greater pregnancy rate up to 150 DIM regardless of first service management. We conclude that a TRM program designed to prioritize AIE by grouping cows based on detection of estrus during the VWP was an effective strategy to submit cows for first service resulting in similar or improved performance than a non-TRM program that prioritized AIE or an all-TAI program with extended VWP. Also, AEA recorded during the VWP might be used as a strategy for identifying subgroups of cows with different reproductive performance.

To determine the effect of the combination of melatonin implants and prostaglandin (PG) F2α on reproductive performance in the late breeding season (Dec at the northern hemisphere), 500 Lacaune ewes were divided into four groups. On day 0 (7 Nov), 150 ewes were treated with a melatonin (M) implant. From that group, 64 ewes (M + 1PGF group) were injected with 10-mg prostaglandin (PG) F2α 34 d after melatonin implantation (11 Dec). The remaining 86 ewes (M group) were treated with melatonin, only. Another group of 75 ewes (2PGF group) was treated with double injection of PGF2α (9 days between the first and second application) (2 and 11 Dec), and 75 non-treated ewes (C group) were the control group. The remaining 200 ewes of the flock were not considered in the study. Rams (n = 23) were introduced on 11 Dec. The percentage of prolificacy, lambing and fecundity rates were calculated. Lambing rate did not differ among groups (M: 79%; M + 1PGF: 78%; 2PGF: 69%; C: 71%). The M + 1PGF group had a higher % of prolificacy than the 2PGF group (P < 0.10) and the C group (P = 0.06) (M: 1.65 ± 0.07; M + 1PGF: 1.74 ± 0.09; 2PGF: 1.54 ± 0.08; C: 1.54 ± 0.07 lambs/lambing) (P < 0.05), and a higher fecundity than the 2PGF group (P < 0.05) and the C group (P < 0.10) (M: 1.30 ± 0.09; M + 1PGF: 1.36 ± 0.11; 2PGF: 1.07 ± 0.10; C: 1.08 ± 0.09 lambs/ewe). Ewes implanted with melatonin had significantly higher prolificacy (1.69 ± 0.06 lambs/lambing) (P < 0.05) and fecundity (1.33 ± 0.07 lambs/ewe) (P = 0.01) than did ewes that did not receive melatonin (1.54 ± 0.04 and 1.08 ± 0.04, resp.). In conclusion, melatonin implants increased the number of lambs born per ewe in a late-autumn mating season, and the effect was greatest if it was given in combination with PGF2α administration at ram introduction.

Changes in the C-reactive protein (CRP) and 13,14-dihydro-15-keto-prostaglandin F (PGFM) concentrations of uterine lavage fluid were examined in cows given an intrauterine povidone-iodine (PI) infusion. The mean polymorphonuclear leukocyte (PMN) ratios (the ratio of PMN to total cells) and CRP concentration of uterine lavage fluid on the day after the treatment were significantly (P<0.05) greater in the PI infusion group (PMN: 53.0 ± 32.7%, CRP: 50.2 ± 32.3 ng/mL) than in the non-treatment control group (PMN: 7.9 ± 21.9%, CRP: 17.2 ± 5.9 ng/mL), whereas there was no significant difference in the mean PGFM concentration between the two groups. The present findings suggest that the uterine CRP level is a useful biomarker of local uterine inflammation in cows.

This study was performed to elucidate whether eicosapentaenoic acid (EPA) suppresses spasm-prone blood vessel contractions induced by a thromboxane mimetic (U46619) and prostaglandin F (PGF) and determine whether the primary target of EPA is the prostanoid TP receptor. Accordingly, we assessed: (1) the tension changes in porcine basilar and coronary arteries, and (2) changes in the Fura-2 (an intracellular Ca indicator) fluorescence intensity ratio at 510 nm elicited by 340/380 nm excitation (F340/380) in 293T cells expressing the human TP receptor (TP-293T cells) and those expressing the human prostanoid FP receptor (FP-293T cells). EPA inhibited both porcine basilar and coronary artery contractions induced by U46619 and PGF in a concentration-dependent manner, but it did not affect the contractions induced by 80 mM KCl. EPA also inhibited the increase in F340/380 induced by U46619 and PGF in TP-293T cells. In contrast, EPA showed only a marginal effect on the increase in F340/380 induced by PGF in FP-293T cells. These findings indicate that EPA strongly suppresses the porcine basilar and coronary artery contractions mediated by TP receptor and that inhibition of TP receptors partly underlies the EPA-induced inhibitory effects on these arterial contractions.

The 8-iso-prostaglandin F2α (8-iso-PGF2α) biomarker is used as the gold standard for tracing lipid oxidative stress in vivo. The analysis of urinary 8-iso-PGF2α is challenging when dealing with trace amounts of 8-iso-PGF2α and the complexity of urine matrixes. A packed-fiber solid-phase extraction (PFSPE)−coupled with HPLC-MS/MS−method, based on polystyrene (PS)-electrospun nanofibers, was developed for the specific determination of 8-iso-PGF2α in urine and compared with other newly developed LC-MS/MS methods. The method, which simultaneously processed 12 samples within 5 min on a self-made semi-automatic array solid-phase extraction processor, was the first to introduce PS-electrospun nanofibers as an adsorbent for the extraction of 8-iso-PGF2α and was successfully applied to real urine samples. After optimizing the PFSPE conditions, good linearity in the range of 0.05−5 ng/mL with R2 > 0.9996 and a satisfactory limit of detection of 0.015 ng/mL were obtained, with good intraday and interday precision (RSD < 10%) and recoveries of 95.3−103.8%. This feasible method is expected to be used for the batch quantitative analysis of urinary 8-iso-PGF2α.

BACKGROUND
Enhanced oxidative stress occurs in atrial fibrillation (AF), however its impact on the efficacy and safety of anticoagulation is unknown. We sought to evaluate whether 8-isoprostaglandin F2 (8-isoprostane) levels are associated with clinical outcomes in anticoagulated AF patients.
METHODS
In a study involving 243 AF patients (median age 69 years), we measured serum 8-isoprostane, along with prothrombotic markers, including plasma fibrin clot permeability, clot lysis time (CLT), endogenous thrombin potential (ETP), von Willebrand factor (VWF), and fibrinolytic proteins. Ischemic cerebrovascular events, major bleeding, and death were recorded during a median follow-up of 53 months while on anticoagulation, largely on non-vitamin K antagonist oral anticoagulants (NOACs).
RESULTS
Increased 8-isoprostane levels were observed in women, in patients with arterial hypertension, and those with paroxysmal or persistent AF. Patients with 8-isoprostane levels ≥559 pg/mL (the top quartile) compared with those with 8-isoprostane <250 pg/mL (the bottom quartile) had higher fibrinogen, lower VWF, higher plasminogen activator inhibitor 1, along with lower fibrin clot permeability with no difference in CHADS-VASc score, CLT or ETP. Patients who experienced thromboembolic events (n = 20, 1.9%/year) had 48.6% higher 8-isoprostane concentrations compared to the remainder (P <0.01). Levels of 8-isoprostane >459 pg/mL based on the optimal cut-off value were associated with thromboembolic events during follow-up (hazard ratio 2.87, 95% confidence interval 1.17-7.03, P = 0.02). There were no associations between 8-isoprostane and major bleeding (2.0%/year) or all-cause mortality (1.9%/year).
CONCLUSIONS
Increased 8-isoprostane levels partly through altered fibrin clot structure are associated with thromboembolic events despite anticoagulant therapy in AF patients.

OBJECTIVE
Renal calculi are a common type of urological calculi and are associated with morbidity. This study was aimed at exploring the effect of flexible ureteroscopy and nephroscopy on stone removal in patients with multiple renal calculi.
METHOD
This randomized controlled trial included a total of 78 cases with multiple renal calculi in our hospital. The patients were randomly divided into the study and control groups and treated with flexible ureteroscopy and percutaneous nephrolithotomy with pneumatic ballistics, respectively. The surgery condition, levels of prostaglandin F2 (PGF2), prostaglandin E2 (PGE2), keratocyte growth factor (KGF), renal function indices, and the incidence of complications were analyzed before and after surgery in the two groups.
RESULT
The operation time, the postoperative analgesia pump application time, one-time stone removal rate, the intraoperative blood loss, and hospital stay of the study group were significantly lower than those of the control group. Postsurgery, the levels of PGE2, PGF2 and KGF in the study group were significantly lower than those in the control group. The serum levels of SCR, BUN, and NGAL in the study group were significantly lower than those in the control group. In addition, the incidence complications in the study group were significantly lower.
CONCLUSION
Flexible ureteroscopy and laser lithotripsy under nephroscopy were equally effective against multiple renal calculi. Flexible ureteroscopy reduced surgical trauma without affecting renal function and had a low incidence of complications.
