Benfuracarb is a carbamate insecticide used to control insect pests in vegetables and it has anti-acetylcholinesterase activity lower than other carbamates. Cytotoxic effects of benfuracarb were evaluated by using root growth inhibition (EC50), mitotic index (MI), and mitotic phase determinations on the root meristem cells of Allium cepa and mutagenic effects were determined in Salmonella typhymurium Ames test by TA98 and TA100 strains with and without metabolic activation. In Allium test, 1 % DMSO was used as negative control group and 10 ppm MMS was used as positive control group. 75 ppm concentration of benfuracarb was found as EC50. In MI and mitotic phases determination study, 37.5, 75 and 150 ppm doses of benfuracarb were used. Dose-dependent cytotoxic activity was found by root growth inhibition and MI studies. It was identified that mitotic inhibition activity of benfuracarb was higher than 10 ppm MMS. In Ames test, mutagenic activity was not observed and over 200 µg/plate of benfuracarb was determined as cytotoxic to S. typhymurium strains. Benfuracarb can be called as "mitotic inhibitor" but not called as mutagen.

To produce a sensitive monoclonal antibody (mAb) for the simultaneous detection of carbofuran, benfuracarb, carbosulfan and 3-hydroxy-carbofuran, 2,3-dihydro-2,2-dimethyl-7-benzofuranmethanamine (DDB) was conjugated to bovine serum albumin (BSA) to prepare the immunogen DDB-BSA and mice were immunized. Coating antigens were prepared by conjugating DDB and 5-methoxy-2,3-dihydrobenzofuran-3-acetic acid (MDA) to BSA and ovalbumin (OVA), respectively. Furthermore, the effect of different antibody-antigen pairs on the sensitivity of ELISA and LFIA methods for the detection of carbofuran was investigated. After the immunization, a high-affinity mAb 13C8 was obtained. The ability of the coating antigen to compete with carbofuran for binding antibodies was found to be significantly different between ELISA and LFIA methods. With the antibody-antigen pair 13C8-MDA-OVA, the IC values of the ELISA and QD-LFIA methods for carbofuran were 0.18 ng/mL and 0.67 ng/mL, respectively. The cross-reactivity (CR) values of the two methods for benfuracarb, carbosulfan and 3-hydroxy-carbofuran ranged from 72.0% to 83.7%, while, for other carbamate pesticides, the CR values were less than 1%. The spiked recoveries of carbofuran in vegetables by the QD-LFIA method were 83-111%, with a coefficient of variation below 10%, and the test results of the actual samples were consistent with the HPLC-MS method. Overall, this study provides key materials for the development of immunoassays for carbofuran and its analogues, and the antibody-antigen pair selection strategy established in this study provides useful insights for the development of sensitive immunoassays for other compounds.

The use of agrochemicals has become a standard practice worldwide to ensure the productivity and quality of sugarcane crops. This study aimed to analyze the metabolic changes in sugarcane culms treated with five different nematicides. The experimental design was randomized in blocks, and agro-industrial and biometric variables were evaluated. The samples were extracted and then analyzed using LC-MS, LC-MS/MS, and LC-HRMS. The data obtained were submitted to statistical methods (PCA and PLS). Fragmentation patterns, retention time, and UV absorptions of the main features were analyzed. The plantations treated with carbosulfan (T4) obtained higher agricultural productivity and total recoverable sugar (TRS), while the use of benfuracarb (T3) was associated with lower growth and lower TRS. Statistical analysis revealed the contribution of the features at / 353 and / 515, assigned as chlorogenic acids, which discriminated the groups. The MS profile also supported the occurrence of flavonoids (C-glycosides and O-glycosides) in the samples.

The antidotal action of atropine sulfate and 2-pyridine aldoxime methiodide (2-PAM) against poisoning attributable to the new procarbamate insecticide benfuracarb [(ethyl N-[2,3-dihydro-2,2-dimethylbenzofuran-7-yloxycarbonyl (methyl) aminothio]-N-isopropyl-beta-alaninate] was compared utilizing rats as our experimental model. Both the intraperitoneal and oral administrations of these antidotes were examined after five, ten, fifteen and thirty minutes exposure periods, following treatment with benfuracarb at dose levels approximating LD50 and LD100. The results obtained demonstrate that both the intraperitoneal and oral administrations of atropine sulfate blocked or significantly reduced the toxic effects of benfuracarb and protected the animals from death. The intraperitoneal administration route appears to be more effective than was the oral route. In addition, the administration of atropine sulfate after the shorter period (up to 15 minutes), following exposure to benfuracarb, improved antidotal action, particularly with the LD100 dose of benfuracarb. It is suggested that atropine sulfate antagonizes benfuracarb poisoning by blocking acetylcholine (ACh) receptors, as many other carbamate insecticides, since benfuracarb was an in vivo cholinesterase (ChE) inhibitor and the toxic effect of benfuracarb was reduced by atropine sulfate. 2-PAM, however, did not significantly block or reduce the toxic effects of benfuracarb.

Cytochrome P450 (CYP) enzymes play a key role in the metabolism of both xenobiotics and endogenous chemicals, and the activity of some CYP isoforms are susceptible to induction and/or inhibition by certain chemicals. As CYP induction/inhibition can bring about significant alterations in the level of exposure to CYP substrates and metabolites, CYP induction/inhibition data is needed for regulatory chemical toxicity hazard assessment. On the basis of available human pharmaceutical data, a draft Organisation for Economic Co-operation and Development Test Guideline (TG) for an CYP HepaRG test method that is capable of detecting the induction of four human CYPs (CYP1A1/1A2, 2B6, and 3A4), has been developed and validated for a set of pharmaceutical proficiency chemicals. However to support TG adoption, further validation data was requested to demonstrate the ability of the test method to also accurately detect CYP induction mediated by industrial and pesticidal chemicals, together with an indication on regulatory uses of the test method. As part of "GOLIATH", a European Union Horizon-2020 funded research project on metabolic disrupting chemical testing approaches, work is underway to generate supplemental validated data for an additional set of chemicals with sufficient diversity to allow for the approval of the guideline. Here we report on the process of proficiency chemical selection based on a targeted literature review, the selection criteria and considerations required for acceptance of proficiency chemical selection for OECD TG development (i.e. structural diversity, range of activity, relevant chemical sectors, global restrictions etc). The following 13 proposed proficiency chemicals were reviewed and selected as a suitable set for use in the additional validation experiments: tebuconazole, benfuracarb, atrazine, cypermethrin, chlorpyrifos, perfluorooctanoic acid, bisphenol A, N,N-diethyl-m-toluamide, benzo-[a]-pyrene, fludioxonil, malathion, triclosan, and caffeine. Illustrations of applications of the test method in relation to endocrine disruption and non-genotoxic carcinogenicity are provided.

BACKGROUND
Pesticide use for fruits and vegetable production in Uganda may result in presence of residues on produce which may pose health risks to consumers. Uganda does not have an established system for monitoring pesticide residues in fruits and vegetables and assessing potential health risks. This research aimed to conduct a health risk assessment of presence of pesticide residues in fruits and vegetables in the Kampala Metropolitan Area in Uganda.
METHOD
Pesticides were measured in 160 fruits and vegetables samples collected at farms, markets, street vendors, restaurants and homes; and analysed using liquid chromatography-tandem mass spectrometry and gas chromatography-mass spectrometry. Fruit and vegetable consumption information was collected from 2177 people. Pesticide concentrations were compared with European Union maximum residual limits (MRLs). Mean values of pesticide concentration residues found in the sample of fruits and vegetables; and fruits and vegetables intake and body weight were used to calculate the estimated daily intake (EDI) of pesticide residues. EDI values were compared with acceptable daily intakes (ADI) to calculate the hazard quotient by age group, and stage at which consumption happens along the chain.
RESULTS
Overall, 57 pesticides were detected in fruits and vegetables from farm to fork. Of the 57, 39 pesticides were detected in all the fruits and vegetables studied. Concentrations of fonofos, fenitrothion and fenhexamid were above the European Union MRLs in some samples. Hazard quotients based on dietary ingestion scenarios for 18 pesticides, including dichlorvos (444) alanycarb (314), fonofos (68), fenitrothion (62), dioxacarb (55) and benfuracarb (24) and others, were above 1, indicating the possibility of chronic health risk to consumers. Chronic health risk decreased with age but was stable for stage at which consumption happens along the food chain. The number of pesticides with EDI greater than the ADI decreased with increase in age; with 18, 13, 9, 11, 8, 9, and 9 pesticides for age groups < 5, 5-12, 13-19, 20-25, 36-49 and ≥ 50 respectively.
CONCLUSION
Chronic dietary pesticide exposures to Ugandans are likely common, and for some pesticides result in exposure exceeding health-based benchmarks. Risks were highest for younger participants. There is an urgent need to increase monitoring and regulation of pesticides in fruits and vegetables in order to protect consumers, especially the children who are vulnerable to the adverse effects of pesticides.

The following N-methylcarbamate pesticides, aldicarb, aldicarb sulfoxide, aldicarb sulfone, oxamyl, methomyl, thiodicarb, propoxur, carbofuran, carbosulfan, benfuracarb, bendiocarb, carbaryl, fenobcarb and furathiocarb, were added to soybean oil, each at 5 mg/kg(5 ppm), followed by degumming, alkali refining, bleaching and deodorization for oil refinement. Residual pesticide content in each case was determined immediately after refining. DEGUMMING: Aldicarb, aldicarb sulfoxide, aldicarb sulfone, oxamyl, thiodicarb, carbosulfan, benfuracarb were each found to decrease by as much as 70% by H(3)PO(4) treatment, this being less than 26% noted for the other pesticides. With hot water treatment, the decrease in any one pesticide was less than 52%. ALKALI REFINING: The rate of decrease varied with the pesticide, ranging from 8% to 100%. 200%NaOH were effectively brought about pesticide removal, compared to 125%NaOH. BLEACHING: Aldicarb, aldicarb sulfoxide, aldicarb sulfone, oxamyl, methomyl, thiodicarb, carbosulfan, benfuracarb, bendiocarb and furathiocarb each decreased by more than 80% with activated clay containing activated charcoal. Carbaryl decreased remarkably by this clay. Pesticide removal in all cases was at less than 30%. DEODORIZATION: 40% Furathiocarb, 14% carbosulfan, 11% benfuracarb and 3% carbofuran could still be detected subsequent to deodorization at 260 degrees C while all other pesticide amounts were too small to permit quantitative detection. Degumming with H(3)PO(4) and bleaching with activated clay caused the conversion of carbosulfan and benfuracarb into carbofuran. Carbofuran and furathiocarb may thus possibly still remain in the oil following the above 4 refinement processes.

Five field trials were conducted in Italy in 1983 and 1984 to test the efficacy of isazofos and benfuracarb in controlling Heterodera carotae on carrot, Ditylenchus dipsaci on onion, and Meloidogyne javanica on tomato. Methyl isothiocyanate (MIT) was tested against H. carotae and M. javanica. Single (10 kg a.i./ha) and split (5 + 5 kg a.i./ha) applications of isazofos gave yield increases of carrot and onion similar to those obtained with DD (300 liters/ha) and aldicarb (10 kg a.i./ha). Population densities of H. carotae in carrot roots at harvest and of M. javanica in tomato roots 2 months after transplanting were also suppressed by isazofos. Benfuracarb (10 kg a.i./ha increased onion yields in a field infested with D. dipsaci, but it was not effective against H. carotae or M. javanica. The efficacy of MIT at 400 and 600 liters/ha was similar to that of MIT + DD (Di-Trapex) at 300 liters/ha. Both nematicides inhibited hatch of H. carotae eggs and decreased the soil population density of M. javanica.

Development of a simultaneous analysis method for carbofuran and its three derivative pesticides in water by GC/MS with temperature programmable inlet on-column injection.
A simultaneous analytical method was examined for carbofuran and its derivative pesticides in water. Since carbofuran derivatives are hydrolyzed to carbofuran in water, the liquid-liquid extraction method was used to obtain an accurate concentration value. Moreover, since these compounds are easily decomposed at the GC/MS injection port, temperature programmable inlet on-column injection was used. By combining the two methods, a sensitive analytical method was established without hydrolysis and thermal decomposition. As a result of recovery experiments using distilled water, river water and tap water, acceptable recovery rates and favorable reproducibility were obtained. This method was used in a field investigation to determine carbofuran and its derivative pesticides in river water taken from three points of the Y river over a period of one year. Carbofuran, benfuracarb, and carbosulfan were detected and corresponded to the period when these pesticides were used in the area. Although benfuracarb and carbosulfan using traditional methods are believed to easily hydrolyze and thermally decompose during the analytical process, by using our method they can be detected.
